Mactra veneriformis is a palatable marine food and a traditional Chinese medicine while being prevalent and affluent in the coastal beaches of Jiangsu province. The bioactive substances for instance polypeptides, lipids, polysaccharides. nucleosides were sought out in Mactra veneriformis. During the previous research process, our teams were dedicated to study the water soluble polysaccharide and polypeptide extracted from Mactra veneriformis. However vast majority of meat residue and shells were obsoleted. For this reason, how to comprehensively utilize Mactra veneriformis meat residue and shells is a particularly significant thing. This thesis is mainly used Mactra veneriformis meal residue and shells as raw materials to research the functional product which has the dual function of supplying calcium and amino acids. Following parts were principally included:1. Literature studyResearch literatures about calcium preparations were referred, and summarized the progress of calcium preparations and the progress of animal models of calcium supplements. Research papers about the methods of enzyme-linked chelating complexing that was used to hydrolyze proteins and to manufacture functional product of amino acid chelating calcium were referred, and the application of marine shellfish biological resources and the methods of enzymolysis proteins using to prepg prepare amino acid were concluded. Thus theoretical basis of further experiment work was established.2. The study of preparation of compound amino add from Mactra veneriformis(1) The enzymolysis of the Mactra veneriformis was studied by measuring the amino nitrogen content and the recovery of nitrogen in the hydrolyzed solution under different conditions. The protein of Mactra veneriformis’ was hydrolyzed by several kinds of proteinase. Contrast experiment was designed to determine the best incision enzyme and excision enzyme of in-outside two steps enzymolysis. The optimized condition of neutral protease and complex protease was chosen by single-factor and orthogonal experiment. The optimal condition was that the 1500U/g neutral protease was added first for the enzymolysis for 6h in the condition of 45℃, pH7.5 and solid-to-liquid 1:3 (w/v), and then 4200U/g complex protease was added for the enzymolysis for 11h in the condition of 45℃, pH7.0 and substrate concentration 1:3 (w/v).(2)The content determinations of free amino acid, total amino acids, total proteins, polysaccharide, fat and metallic element were measured in Mactra veneriformis. Results showed that the effect of two-step enzymolysis was superior to single-step enzymatic hydrolysis.3. The study of preparation of compound amino acid chelate calcium from Mactra veneriformis(1) Optimized the compound amino acid chelate calcium process conditions that was prepared by compound amino acid and shell powder from Mactra veneriformis, the best chelation process conditions are as follows:compound amino acid and calcined shell powder mass ratio was 2:1, adjusted to pH 6.0,60 ℃ water bathing for 1.5 hours. The analysis of UV scanning and infrared scanning had proved that compound amino acid chelate calcium formation.(2) Further tests of composition analysis on elemental calcium, amino acid, total protein, the water content, ash content and extractable metal content were conducted. Quality standard of compound amino acid chelate calcium from Mactra veneriformis was built by optimizing the measurement method.4. The study on acute toxicity test and treatment of osteoporosis in rats by amino acid chelate calcium from Mactra veneriformis(1) Further test is acute toxicity test of amino acid chelate calcium from Mactra veneriformis. The results showed that normal diet and activity, the growth and development in mice were favorable, and the mice did not see obvious poisoning symptoms and no death phenomenon. According to acute toxicity classification standard, this chelating calcium belongs to actually non-toxic substance(2) The effects of calcium supplementation experiment of amino acid chelate calcium from Mactra veneriformis proceeded by using the animal model of osteoporosis rats induced by retinoic acid. In this experiment, general physical sign was observed, metabolic test indicators, serum biochemical indices and femoral growth indicators were determined. The results showed that amino acid chelate calcium from Mactra veneriformis has obvious therapeutic action about retinoic acid induced osteoporosis. |
