Objective:To investigate the relationship between drug-resistant SGC7901 cells and Runx3 gene.Methods:Drug-resistant cell SGC7901/cisplatin(CDDP) dynamics model SGC7901/CDDP-1.0 was established,and then treated with 5-Aza-2’-deoxycytidine(5-Aza-CdR).SGC7901/CDDP-0.8 was transfected with Runx3 interference Plasmid.Cell proliferation activity,the cell cycle distribution and apoptosi were measured by CCK-8 assay,Flow cytometry (FCM) and Hoechst 33258 respectively.Reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting were used to determine Runx3 mRNA and the expression levels of P-gp and Runx3.Results:in SGC7901/CDDP-1.0 dynamic model with CDDP,Runx3 mRNA expression gradually reduced. After treated 5-Aza-CdR,the expression of Runx3 mRNA increased,cell growth was significantly inhibited,S phase cells increased gradually,G1 phase cells gradually reduce, apoptosis significantly increased,reduced P-gp expression.After transfection,The tolerance of CDDP for SGC7901/CDDP-0.8 significantly enhanced,and the expression of Runx3 mRNA and protein reduced.Gl phase cells increased, the expression of P-gp increased in the cells.Conclusion:Runx3 gene in the formation of drug resistance of SGC7901 cells has the vital role.
