BACKGROUD:This study aimed to observated the effect of GLP-1 on high glucose-induced cell proliferation and apotosis in rabbit thoracic aortic endothelial cells(RTAECs) and HSP70 expression.Methods: RTAECs were isolated from rabbit thoracic artery and cultured under varying different dictions. Tunnel was used to assay endothelial cells apoptosis. Cell proliferation was assayed with BUDR. The protein lever of HSP70 were detected by western blot.Results: Compared with the control group, the apoptosis and the protein level of HSP70 of RTAECs cultured under high glucose(33m M) for 24 hours increased, but proliferation was inhibited(p﹤0.05). Pretreatment with GLP-1(10﹣8M) in the high glucose group could decrease the apoptosis ratio and HSP70 expression and promote proliferation of RTAECs(p﹤0.05). The effect of GLP-1 on the RTAECs was inhibited by exendin(9–39)(10﹣7M)(GLP-1 receptor antagonis). Compared with the high glucose group, there wasn’t significant difference in the exendin(9–39) group about the apoptosis, proliferation and HSP70 expression of RTAECs.Conclusion: GLP-1 can ameliorate high-glucose-induced RTAECs apoptosis and promote cell proliferation, and attenuate high-glucose-induced the level of HSP70 expression through the GLP-1R-dependent pathways. The protective effect of GLP-1 on vascular damage by high glucose is probably related to lower expression of HSP70.
