Objective:Bile duct carcinoma is associated with inflammation of the bile duct tumor, NF-κB signaling pathway is one of the numerous chain of "inflammatory and cancer", CUEDC2 is a kind of cohesion in protein helps to inhibit the NF-κB signaling pathway, and CUEDC2 is overexpression in a variety of malignant tumors, but in cholangiocarcinoma little studied.1. The relationship between the expression about CUEDC2, p-IKKβ, NF-κB innormal bile duct, bile duct stones, bile duct carcinoma and the clinical pathologicalcharacteristics will be discussed;2. Explore the relationship between NF-κB signaling pathway and bile ductcancer;3. Provide a preliminary experimental basis for the diagnosis and prognosis ofbile duct cancer.Methods:The study cases were collected in hepatobiliary surgery ward in our hospital during June 2011 to September 2014. Surgical diagnosis of bile duct cancer patients, 61 cases, as bile duct cancer group, and over the same period, patients with bile duct stones 22 cases as gallstone group, other resected specimens in 18 cases of normal bile duct as control group.They were recorded in detail, the general information and clinical pathological information were collected.I detected the expression of CUEDC2, p-IKKβ, NF-κB respectively among the three kinds of tissue by using the method of immunohistochemistry;Analysis of the clinical characteristics, pathological features and the correlation between the three kinds of molecular expression of the stone and bile duct cancer;Semi quantitative analysis ofCUEDC2 and TNF-α in the three groups by Western bolt; The results were statistically analyzed by using the thirteenth version of SPSS statistical software.Results:CUEDC2 protein expression rate in extrahepatic bile duct stones, bile duct carcinoma and adjacent tissues were 6%, 36% and 87%, compared with the three groups, the bile duct cancer group was significantly higher than the other two groups,P < 0.05 had statistical significance; p-IKKβ expression rate in the three groups of specimens were 17%,59% and 82%, the bile duct cancer group were significantly different from the other two groups, P < 0.05; NF-κB positive rate in the three groups of specimens were as follows:11 %, 50%, 85%, compared with normal bile duct and bile duct stones, the expression of NF-κB in bile duct cancer group was significantly higher than that in normal bile duct carcinoma, and the difference was statistically significant.CUEDC2, p-IKKβ, NF-κB expression difference has no correlation wiht bile duct cancer patients gender, age, tumor diameter(P > 0.05), and closely related to tumor cell differentiation degree, lymph node metastasis, infiltration of blood vessels nerves, and TNM staging(P< 0.05); The expression level of CUEDC2 was correlated with the length of chronic inflammation of the bile duct carcinoma(P < 0.05); Correlation analysis, three groups of specimens of p-IKKβ, NF-κB expression was positively correlated(r = 0.876,r=0.543, r = 0.491). CUEDC2 in normal bile duct group respectively and p-IKKβ, NF-κB expression was positively related(r=0.257, r = 0.414);The expression of CUEDC2 in bile duct stones was negatively correlated with p-IKKβand NF-κB(r=-0.311).In stone group and cancer group, CUEDC2 and inflammatory cytokines TNF-αexpression significantly statistical difference, P< 0.05, and positively correlated; The expression level of CUEDC2 protein in simple inflammatory hyperplasia group is higher than atypical inflammatory hyperplasia group(0.455 + 0.03) /(0.197 + 0.02), P < 0.05],TNF-α expression in simple inflammatory hyperplasia below atypical inflammatory hyperplasia group(0.289 + 0.01) /(0.500 + 0.02), P < 0.05], the difference is statistically significant.Conclusion:CUEDC2, p-IKKβ, NF-κB three molecules in cholangiocarcinoma tissues showed high expression. CUEDC2, p-IKK beta, NF-κB high expression in bile duct carcinoma has relationship with the degree of tumor differentiation, tumor invasion, TNM staging; There is a close relationship between NF-κB signaling pathway activation and bile duct cancer.In the development of cancer, the amount of CUEDC2 increased with the increase of the levels of inflammatory factors TNF-α, and the expression of CUEDC2 inhibited the conversion of inflammatory hyperplasia to atypical hyperplasia.
