Objective:The use of experimental rabbits oral tissue + complete Freund’s adjuvant injection method to establish an animal model of recurrent oral ulcers, by detecting recurrent oral ulcers(Recurrent oral ulcers, ROU) rabbit serum interleukin-6(Interleukin-6, IL-6),interleukin-8(Interleukin-8, IL-8) expression to study the effect of vitamin B12 on recurrent oral ulcers lesions.Methods:57 New Zealand rabbits, laboratory adaptive feeding week. Acute sudden death among 15 to take the oral mucosa backup, and the remaining 42 rabbits were randomly divided into 2 groups, namely the control group(11), model group(31). After modeling 12 weeks, according to the random number method, a randomly selected control group, the experimental group of rabbits each one, cut sudden death after oral antigen preparation.After successful modeling, the model group, 30 were randomly divided into group B(positive control group), group C(levamisole group), D group(vitamin B12 group). A group(control group) and group B after model using distilled water, in group C levamisole tablets orally, D group with vitamin B12 intragastrically administered for two weeks,observed during the recording of oral ulcers in rabbits changes after the gavage histopathology after HE staining light microscope; ear vein blood using enzyme-linked immunosorbent assay after each group of rabbits fed the end of serum IL-6, and IL-8content.Results:1. B, C, D group to the oral mucosa tissue + complete Freund’s adjuvant as the experimental group antigens are both rabbits with oral mucosal hyperemia, some erosion occurs, serum anti-antibody showed positive for the oral mucosa. After the tissue HE staining light microscope, showing the continuity of epithelial damage, intraepithelial dense infiltration of inflammatory cells, lamina propria lymphocytes visible, neutrophil infiltration, capillary congestion expansion, partially visible gland hyperplasia. ROU model rabbit body weight and food intake were significantly different from the process of change in group A. B, C, D group was given after the end of each group cut ulcers, routine HE staining, pathology observed in group B undermine the continuity of the mucosal surface,and there is pseudo-embranous covered with dense intraepithelial lymphocytes, monocytes cell infiltration, a large number of inflammatory cells in the lamina propria infiltration,capillary hyperemia, vascular endothelial cell swelling, partially visible necrosis. Group C and Group D healing of ulcers, thickening of epithelial integrity, submucosal for loose connective tissue, showing new capillaries.2.serum showed that, after the gavage, group B and group A, B of IL-6, IL-8 were significantly increased, p<0.01, a statistically significant difference; group C and D and B group, two groups of IL-6, IL-8 levels were significantly lower than the positive control group, p<0.01, a statistically significant difference; D group compared with group C, the difference was not statistically significant.Conclusions:1. the use of rabbit oral mucosal tissue homogenates as antigens, can cause the occurrence of experimental rabbits ROU, its clinical manifestations and histological examination were similar to human ROU, provide an experimental basis for further study of recurrent oral ulcers;2.vitamin B12 through micronutrient supplementation, regulate immune function,expression and regulation of cytokines in the body, a significant reduction in serum IL-6,IL-8 high-level content, which may be the treatment of recurrent oral ulcers mechanism for one.
