Objective:By feeding high-fat diet SD male rats as the research object, observation the role and significance of lentivirus mediated CMKLR1 expression in adipose tissue of SD rats with non-alcoholic steatohepatitis.Methods:Made in advance with rats CMKLR1 gene lentivirus vectors construction and evaluation.42 male SD rats,in animals adapt to raise after 7 days, all the rats were divided into four groups at random:(1)reference only 12;(2) model 12;(3) transfection 12;(4)empty transfection 6, control group normal diet;The rest of the high fat diet feeding.In experiment 1 day, control and model groups into tail vein injection PBS 100 ul,transfection group in caudal vein injection disease venom 100 ul, empty transfection group into tail vein injection empty disease venom 100 ul.The experiment 7, 14, 20 sky transfection group and 2 of the execution, evaluate transfection efficiency.Weekly record weight of rats;Experimental 8, 12 weeks, each group randomly take 6 mice in the diet after12 hours, chloral hydrate anesthesia, abdominal cavity quickly cut open the abdominal cavity, observe the liver morphology, abdominal aorta in blood, kidney week adipose tissue,frozen in liquid nitrogen, and then transferred to- 80 ℃ refrigerator frozen storage, save the liver HE staining, kay Murray, adiponectin levels in the blood;Rt-pcr method for detecting CMKLR1 in the fat tissue, the content of adiponectin m RNA;Protein immunoblot method of measuring each CMKLR1 in adipose tissue of rats and the protein content of adiponectin.Results:(1) model rats control over the same period and transfection group rats weight and liver index increased significantly, the difference was statistically significant(P < 0.05).(2)HE liver pathological results showed that the control rat liver cells in a certain order, livercell line structure clear, radial arrangement;Eight weeks of model group rats appear simple fatty liver, view inside the liver cell swelling, disorganized Gan Tiao and hepatic sinus disappeared, liver cells surrounding the lobules appear focal fatty degeneration;Within 12 weeks of severe fatty liver, vision appear almost all of the liver cells is given priority to with big bubble sex fat drops of mixed fatty change, and inflammatory cells infiltration and dot necrosis, progress for NASH;Transfection group 8 weeks have appeared in the simple fatty liver, NASH has also appeared in 12 weeks, but the degree is lighter than the same period model group.(3) the model group than the control group rats serum kay Murray levels rise, but there was no statistically significant difference, transfection group than the same period model kay Murray levels rising, the difference was statistically significant(P< 0.05).(4) the model group than the control group rats serum adiponectin levels;Transfection group of adiponectin levels in the model over the same group, the difference had statistical significance(P < 0.05).(5) the rt-pcr results show that the model group compared with the same period the fat tissue of rats CMKLR1 m RNA expression increased, but there was no statistically significant difference, transfection group and model group significantly higher than the same period, the difference was statistically significant(P < 0.05);Compared to model group compared with control group rats reduced adiponectin m RNA expression in adipose tissue, transfection group and model group significantly higher than the same period(P < 0.05), the difference had statistical significance.(6)group and model group significantly higher than the same period, the difference was statistically significant(P < 0.05);Model group compared with the same period the fat tissue of rats in adiponectin protein content decreased, transfection group and model group significantly higher than the same period, the differences were statistically significant(P < 0.05).Conclusions:A high-fat diet feeding success established the SD rats nonalcoholic fatty hepatitis model; The lentivirus mediated CMKLR1 over expression in adipose tissue of rats can significantly improve the SD rats with nonalcoholic fatty hepatitis liver tissue pathology change.
