| Objective:To expore the expression of ACE2 m RNAã€Mas receptor m RNA and IGFBP-2 m RNA of hepatic stellate cells,so we cultivated hepatic stellate cells in vitro to confirm relation of ACE2- Ang(1-7)- Mas axis and hepatic fibrosis from molecular level and provided the theory basis for the diagnosis and treatment of liver fibrosis.Methods:HSC-T6(rat hepatic stelate cell) were cultured in 37 ℃ and 5% CO2 conditions, in containing 10% fetal bovine serum, 100 u/ml/penicillin streptomycin 1640 medium, to extend a week 2 or 3 times. HSC-T6 were randomized into five groups:1.The control group 2.Angâ…¡group 3.Ang(1-7) group 4.Angâ…¡+Ang(1-7) group 5.Ang II+Ang(1-7)+A779(group for Mas receptor antagonists).when they cultured 24 hours, and then cracked cells, extractd total RNA, to reverse transcription, detection the expression of ACE2 m RNA, Mas receptor m RNA and IGFBP-2m RNA by real-time fluorescent quantitative PCR.Results:(1) Compare to control group,the expression of ACE2 m RNAã€Mas receptor m RNA and IGFBP-2 m RNA in Angâ…¡ group are raised(P<0.05)(2) ACE2 m RNA and Mas receptor m RNA are increased in Angâ…¡+ Ang(1-7) group than Angâ…¡group(P <0.05)IGFBP-2m RNA are decreased in Angâ…¡+Ang(1-7) group than Angâ…¡group(P<0.05)(3)Compared with Angâ…¡ group, expression of ACE2 m RNA, Mas receptor m RNA,IGFBP-2m RNA in Angâ…¡+Ang(1-7)+A779 treatment group have no statistical difference(P >0.05).Conclusions:(1) There are expression of ACE2,Mas receptor and IGFBP-2 in hepatic stellate cells(2) Ang(1-7) in the activation of HSC can raise expression of ACE2 m RNA and Mas receptor m RNA and weaken IGFBP-2 m RNA expression, Mas receptor blocker A779 could block Ang-(1-7) the effect of antagonism Angâ…¡(3) ACE2 m RNA,Mas receptor m RNA and IGFBP-2m RNA were increased than Angâ…¡group, indication of ACE2, Mas receptor and IGFBP-2 is partaked in liver fibrosis. |
