Objective: To investigate the effect of vitamin A on the function of Th17 and Treg cells in the peripheral blood of asthmatic children, and to investigate the mechanism of immune regulation by vitamin A.Methods:we Selected 20 asthmatic children as the object of study who received treatment in the Affiliated Hospital of Medical College, Qingdao University Medical hospital form September 2014 to January 2015,healthy children and for hydrocele of tunica vaginalis or indirect inguinal hernia hospitalized children 16 cases as the control group. Density gradient centrifugation was used to obtain suspension of peripheral blood mononuclear cells(PBMC),then they were given different concentrations of vitamin A preparations(0 mol / L, 0.5 mol / L, 1 mol / L, 2 mol / L) processing In vitro, cultured for 72 hours, left the supernatant, enzyme-linked immune adsorption assay(ELISA)was used to determine the IL-17, IL-10 and TGF- 1 concentration in the supernatant. The comparison between the two groups was t test, and the single factor analysis of variance and LSD-t test were compared among the groups..Results: ①IL-17 level produced by PBMC in the asthma group were significantly higher than that in the normal control group, and the levels of IL-10 and TGF-β1 were significantly lower than that in healthy control group, and all the differences have statistical significance(P < 0.01). ②IL-17 level produced by PBMC in the low, medium and high vitamin A concentration of the asthma group were significantly lower than that in blank control group, and IL-17 level in the high vitamin A concentration were significantly higher than that in low and medium concentration group, and all the differences have statistical significance(P < 0.01). IL-17 level in the medium vitamin A concentration were lower than that in low concentration group, but the difference was not significant(P > 0.05). ③ IL-10 level produced by PBMC in the low, medium and high vitamin A concentration of the asthma group were significantly higher than that in blank control group, and IL-10 level in the low and medium vitamin A concentration were significantly higher than that in high concentration group, and all the differences have statistical significance(P < 0.01). The difference of IL-10 level in the low and medium vitamin A concentration was no statistical significance(P > 0.05). ④ TGF-β1 level produced by PBMC in the low, medium and high vitamin A concentration of the asthma group were significantly higher than that in blank control group, and TGF-β1 level in the low and medium vitamin A concentration were significantly higher than that in high concentration group, and TGF-β1 level in the medium vitamin A concentration were significantly higher than that in low concentration group, and all the differences have statistical significance(P < 0.01).Conclusion:①The function of Th17 in Children with asthma during asthma attack are enhanced, and the function of Treg cells are reduced. The balance of the functions of Th17 and Treg cells occur disorder. ② Vitamin A can reduce the function of Th17 in peripheral blood, and enhance the activity of Treg cells in children with asthma. The physiological level of vitamin A has the best effect, this effect is decreased significantly by high vitamin A concentration.
