Inflammation caused by the implanted biomaterials is regulated by the structure and performance of the biological materials, such as the structure and morphology, the physical or chemical signalsand the biological information of the biomaterials surface. At the same time, these performances of the biological materials have important influences on the biology behaviers of endothelial cells(proliferation, differentiation and release of inflammatory factor, etc.).Random and ordered poly-L-lactic acid(PLLA) fibers were prepared with different diameters(380 nm, 980 nm, 1500 nm) by controlling the electrostatic spinning parameters. Following, the PLLA fibers were surface modified by layer-by-layer(LBL) self-assembling with chitosan, heparin and graphene oxide. Also, surface modification was developed via self-polymerization of biological adhesive dopamine and covalent graft of fibronectin.The physical and chemical properties of functional PLLA fibers surface was characterized by scanning electron microscope(SEM), X-ray photoelectron spectroscopy(XPS) and Fourier transform infrared spectroscopy(FTIR). And then, cells morphology, cell proliferation, cell cycle as well as the release of inflammatory factorswere carried out to researchthe influence of the structure and properties of PLLA fibers on endothelial cells.The experimentsturn out that: 380 nm PLLA fibers could promote the growth and proliferation of the endothelial cells, which shows weaker inflammatory response. Compared with the PLLA fibers, PLLA-CS/Hep fibers and PLLA-CS/Hep/GO fibers have improved the proliferation activity of endothelial cell, and PLLA-CS/Hep fibers have inhibited the inflammatory response in a way. PLLA-DAP/Fn fibers have improved the activity of endothelial cell proliferation, and promote the release of TNF- a and IL-1β.
