Objective: By measuring Sessile minimum inhibitory concentration(SMIC) and its adhesion effect of Tuoguan San resisting biofilm and observing the bio-film form and c test, we discuss the mechanism of Tuoguan San resisting bio-film of pseudomonas aeruginosa.Method: Applying the plate method to cultivate the biofilm of pseudomonasaerugino sa and XTT reduction method to detect and evaluate biofilm adhesion effect of Tuogu an San resisting pseudomonas aeruginosa and its SMIC, we canobserve the influence of Tuoguan San on pseudomonas aeruginosa biofilm form under the electron microsco pe. Also, we can detect its hemolysis effect.Result: 2000μg/m L and 200μg/m L of Tuoguan San resist biofilm adhesion of PA remarkable for 1hour,2hours,4hours.The resistance effect better, the time was lon ger at the same concentration. The effect of Tuoguan San resisting bio-film adhesion of PA had a dose-effect and time-effect relationships. The SMIC50 and SMIC80 were 80μg/m L and 175μg/ml. Comparing with the blank controlgroup, the biofilm were chan ging from thick and sticky to little, thin and scat-tered, that are obvious under the SE M. This changing were obvious along withthe drug concentration increased. When the concentration of Tuoguan San was 3200μg/m L, the percentage of hemolysis was only 3.89%.Conclusion: Effective dose of TuoguanSan can significantly suppress early development pseudomonas aeruginosa biofilm and damage the form of biofilm, which have strong effect to clear the biofilm. That Tuoguan San damage the Quorum-sensing of the biofilm was possibly the mechanism and the hemolysis of Tuoguan San was not happened significantly.
