Objective Postmenopausal osteoporosis is a comprehensive disease and characterized by low bone mineral density, and mainly base on estrogen decline. treatment of women with hormone replacement therapy can prevent bone loss. Phytoestrogens estrogenic effects and little toxicity or side effects can prevent bone loss and Maintain normal bone metabolism. Compared with western countries’ low dietary phytoestrogens consumption, the incidence of Postmenopausal osteoporosis still not decreases in our country. Thus, we suspected that genetic differences among individuals played a role. In the past our group have find that the ERα gene polymorphisms were releared with the dozes of dietary intake of phytoestrogens and Postmenopausal osteoporosis. But There is no report about the relationship between the effect of phytoestrogens anesising Postmenopausal osteoporosis and ERβ gene polymorphisms until now.So we planed to combine with estrogen receptor beta gene polymorphism and dietary intake of phytoestrogens to analyze their relationship while they effect Bone metabolism, and provide a theoretical basis for further research treatment.Methods we randomly selected 300 postmenopausal women from Nan Cha ng,according to the requirement of the investigation, collected basic material a nd diatery material of the samples.BMD of different bony tissues were measur ed by dual energy X-ray absorptiometry. After calculating the ADI(acceptable d aily intake) of all kinds of phytoestrogen,we make a comparison about charac teristics and BMD according to tertiles of phytoestrogen intake. What’ more,Th e Rsa I and Alu I polymorphisms of ERβ gene were detected by Polymerase ch ain reaction-restriction fragment length Polymorphism(PCR-RFLP).Results(1)The differences of BMD of lumbar spine were significant with high, medium and low groups with dietary phytoestrogens intake dose.(2)The result of correlation analysis showed that RR,Rr,rr genotypic freq uence had correlation with BMD of L4(p<0.05).(3) The result of correlatio n analysis showed that there was not statistacally siginificant difference betwee n effect of dietary phytoestrogens on bone mineral density and diverse genetyp es of Alu I and Rsa I(p>0.05).Conclusions( 1) The result of correlation analysis showed that the polymorphisms of Alu I had no correlation with BMD of all the bone tissue of postmenopausal women.(p>0.05).(2) The result of correlation analysis showed that RR genotype had correlation with BMD of lumbar spine(p<0.05).(3) We found no relationships between the ERβgene polymorphisms and effect of dietary phytoestrogens on bone mineral density in postmenopausal women of Nanchang.Objective To observe the effects of GEN(a kind phytoestrogen)on osteoclasts differentiation from RAW264.7 cells, and osteoblasts differentiation from MC3T3-E1 cells,and investigate which estrogen receptor the GEN combined, then it provide the bone-protective function to inhibit the effect that RANKL and MCSF induce RAW264.7 cells to differentiate to mature osteoclasts, and promte the MC3T3-E1 cells differentiation to mature osteoblasts.Methods Osteoclasts were induced by 50ng/ml RANKL and 25ng/ml MC SF from RAW264.7 cells in vitro, and treated with 1×10-9 mol/l、1×10-8mol/l、1×10-7mol/ml GEN, MPP(ERαantagonist), PHTPP(ERβantagonist).When ost eoclasts maturated, TRAP staining and bone resorption were used to demonstra te the differentiation of osteoclsts. meanwhile, we induced the MC3T3-E1 cells to differentiate to mutureated osteoblasts with suitable concentration Vc,β-gly cerophosphate, treat measurement is same with raw264.7,after culturing,adopted ALP staining and mineralized bone nodular structure to observe how the GE N regulated the function of osteoblasts differentiation by combing with either ERα or ERβ.Results The dose of GEN ranged from 1×10-9mol/l to 1×10-7mol/l,we found the 10-8mol/l GEN possessed the most effective function to promote the MC3T3-E1 differentiate to the mature ones,the MPP and PHTPP could offered the effect of down-regulation of the number of ALP positive cells.we found that all these situation and tendency also presented among the experiment of mineralized bone nodular structure.Conclusions In the process of osteoblast materialism,the ability of osteoblast differentiation expressed a tendency that increased at first and then decreased with the dose of GEN ascending.Both MPP and PHTPP could inhibit the promotion-function of GEN. Meanwhile in the process of osteoclast materialism,GEN resisted the differentiation and multiplication of osteoclast, even the antagonism effect was related with dose closely.The ability of osteoclast differentiation had down-regulated with ERαbeing blocked by MPP,however the similar phenomenon didn’t appear among ERβ.
