Objective: To investigate the influence of hydrogen sulfide through PI3K/Akt signal on the liver of rats with experimental hepatic fibrosis by observing the expression of Bad and GSK-3β. Method: The hepatic fibrosis rat models were established by hypodermic injection of carbon tetrachloride mixed with cottonseed oil at the concentration of 40%, feeding high-fat, high-cholesterol diet and drinking ethanol. The rats were randomly divided into five groups after six weeks: hepatic cirrosis group(group C), DMSO group(group D), LY294002 group(group LY), Na HS group(group S), and LY294002+Na HS group(group LS). The rats in group C, group D, group LY and group S were intraperitoneally infused with physiologic saline, 2‰ DMSO solution, LY294002 solution(0.3mg/kg·d), and Na HS solution(56μmol/kg·d) separately for 12 times. The rats in group LS were intraperitoneally infused with LY294002 solution(0.3mg/kg·d) and Na HS solution(56μmol/kg·d) simultaneously for 12 times. All rat livers were collected after all above treatments were finished. Hepatic fibrosis pathology stages were determined by HE staining. The depositions of collagen fiber were observed by Masson staining. The expressions of Bad and GSK-3β were tested by western blot. Result:(1)HE staining:C group of the liver lobule structure disorder, fibrous connective tissue hyperplasia, false flocculus to form, degeneration necrosis of liver cells and inflammatory cells infiltration.(2)Masson staining : C group of hepatic lobule structure destruction, collagen fibers deposition in great quantities, fiber interval increased, broadening the density, is a typical false flocculus structure.(3)Compared to the group N and group S, the expression of Bad and GSK-3β in the group C and group D were over expressed; Compared to the group LS,the expression of Bad and GSK-3β in the group LY was increased and that in the group S was decreased. Conclusion:(1)Through PI3K/Akt signal,Hydrogen sulfide suppresses the expression of Bad and GSK-3β and alleviates the liver fibrosis of rats with experimental hepatic fibrosis.
