Objective: Optimize the extraction condition of Anoectochilus roxburghii, separation and purification the chemical constituent of Anoectochilus roxburghii by using the adsorption material such as silica gel, to obtain objective constituent FAB and other chemical constituents of Anoectochilus roxburghii, their structures were identified by the spectroscopic method such as IR、MS、1D-NMR and 2D-NMR.Methods: 1. Using the high performance liquid chromatography, the peak area of FAB was used as extraction metrics, static extraction times and flushing volume was investigated, to design single factor and orthogonal scheme and determine the optimal extraction conditions of FAB.2. In view of the different extraction parts, separation and purification of repeatedly the different constituent by using the adsorption material such as positive phase silica gel, polyamide, macroporous adsorption resin and ODS, coupling with preparative high performance liquid chromatography for further purification and collection the different constituent.3. The spectroscopic method such as IR、MS、1D-NMR and 2D-NMR were used to identify the structure of compounds which were obtained.Results: 1. The optimal method of extraction Anoectochilus roxburghii by using accelerated solvent extraction was as follows: 85℃, static extraction for 3 times, each time 8 min, 100% flush volume.2. After separation and purification the chemical constituent of Anoectochilus roxburghii by using the adsorption material such as silica gel, gave 21 compounds. 20 of these compounds were identified by the spectroscopic method such as IR、MS、1D-NMR and 2D-NMR, After identify, these 20 compounds are as follow:(22E, 24ξ)-24-isopropenyl-22-dehydrocholesterol(1), Anoectosterol(2), Stigmasterol(3), β-sitosterol(4), Hircinol(5), Moscatin(6), 5,3′-dyhydroxy-3,7,4′-trimethoxyflavone (7), Linoleicacid(8), 8,9-epoxyheptadeca-1,11,14-triene(9), acetovanillone(10), parahydroxybenzaldehyde(11), parahydroxyacetophenone(12), bis(2-ethylhexyl) phthalate(13),7,3′,4′–trimethoxy-quercetin-3-O-α-L-rhamanopyranosyl(1→2)-β-D-gluc opyranoside(14),3′–methoxy-quercetin-3-O-α-L-rhamanopyranosyl(1→2)-β-D-glucop yranoside(15), β-Adenosine(16), isorhamnetin-3-O-β-gentiobioside(17), Loroglossin(18), corchoinoside C(19), parahydroxybenzylalcohol(20).Conclusion: Compound 1, 5, 17, 20 were isolated from orchidaceae for the first time; Compound 6, 7, 10, 12, 13 were isolated from anoectochilus for the first time; Compound 9, 14, 16, 19 were isolated from this plant for the first time.
