The study will further reveal the dermal stem cells function in the process of embryonic development and explore the mechanism of action in wound healing.Methods:Skin tissue dermal stem cells will be isolated from mouse embryos through the identification of cell surface markers.Using immune magnetic bead separation technology,the DLK1+/Sca1-and DLK1-/Sca1+mesenchymal stem cells will be sorted out and the supernatant will be collected.DLK1+/Sca1-cells will be induced by multi-directional differentiation.The wound healing activity of DLK1+/Sca1- cells will be investigated by excision and incision wound healing models.Results:1. Dermal-derive stem cells by flow cytometry instrument positive expressed CD29(mesenchymal stem cell surface markers), while negative expressed CD34.2. Use the immune magnetic bead separation technology to sort of DLK1+/Sca1-and DLK1-/Sca1+ cell.DLK1+/Sca1- cell,thorough cell immunofluorescence technique, found that the positive expression of CD90, CD44.Inducting bone/fat differentiation medium,DLK1+/Sca1- and DLK1-/Sca1+ can be differentied into the direction of osteogenesis/fat cells.3.
