| ObjectiveDetecting the expression of Trk B and BDNF in the cervical lesions and the infection status of high-risk human papillomavirus at the same time. In order to explore the role of these three factors and their relationships in the development of cervical squamous cell carcinoma.Methods1. Selected 106 cases of CIN and squamous cell carcinoma tissue as the study group,including 71 cases of cervical intraepithelial neoplasia(20 of CIN â… ,28 of CIN â…¡,23 of CINâ…¢),and 35 cases of cervical squamous cell carcinoma;Also selected 20 cases of normal cervical tissue as controls.2.All 126 specimens from the cervical canal were collected for HPV-DNA infection status and the genetic type of HPV-DNA detection using Flow-through hybridization and gene chip(Hybri Max).3.Using the Two-steps Method of Immunohistochemistry to detect the Trk B and BDNF in those five groups, and Then using the Image-Pro Plus 6.0 image analysis system to measure the intensity of Trk B and BDNF expression in a form of integrate optic density(IOD)with mean±standard deviation.4.Using SPSS13.0 statistical software to analyze the results.Results1.The proportion of High-risk HPV in the normal cervical tissue,CIN I,CIN II,CIN III and cervical squamous cell carcinoma,respectively 5.00%ã€60.00%ã€75.00%ã€86.95%ã€94.28%,and the trend was gradually increasing.The most three common subtype of HPV infection in the study were HPV16,58,33.2.The IOD of Trk B positive expression in the controls, CINâ… ,CINâ…¡,CINâ…¢ and cervical squamous cell carcinoma were 1.775±0.499ã€3.932±0.774ã€7.223±1.469ã€11.355±2.111 and17.102±2.421.Such differences among these five groups was statistically remarkable(P﹤0.05).The expression strength of Trk B significantly increased with the rising of the degrees of cervical lesions.The IOD of BDNF positive expression in the controls, CINâ… ,CINâ…¡,CINâ…¢ and cervical squamous cell carcinoma were1.653±0.575 〠3.124±0.730 〠5.013±0.761 〠8.688±1.773 ã€16.524±2.773.Such differences among these five groups was statistically remarkable(P﹤0.05).The expression strength of BDNF significantly increased with higher degrees of cervical lesions.The IOD of BDNF and Trk B positive expression in cervical squamous cell carcinoma were17.102±2.421ã€16.524±2.773。They positive correlated with each other,γ=0.899,P<0.05。3.The explession strength of Trk B and BDNF increased gradually with the increase of clinical stage of cervical squamous cell carcinoma(P<0.05).The expression of Trk B and BDNF in poorly differentiated group were significantly higher than moderately differentiated and high group(P <0.05).4.The expression of Trk B and BDNF in HR-HPV group were significantly higher than LR-HPV and negative group(P <0.05).There were no difference between LR-HPV and negative group.5.In the group of the controls,CINS and cervical squamous cell carcinoma groups,the expression of Trk B and BDNF of high-risk HPV higher than that of HPV-negative,the difference was statistically significant. According to bivariate correlation analysis,in cervical lesions,Trk B and BDNF both positive correlated with high-risk HPV infection.6.In the group of the controls,CINS and cervical squamous cell carcinoma groups,the expression of Trk B and BDNF of high-risk HPV higher than that of HPV-negative,the difference was statistically significant. According to bivariate correlation analysis,in cervical lesions,Trk B and BDNF both positive correlated with high-risk HPV infection.Conclusion1, High-risk HPV infection is correlated with the progression of CIN and cevical cancer.2,The expression of Trk B and BDNF can be seen in controls,CINS,and cervical cancer.However the expression strength were increased with higher degrees of cervical lesions,which indicated that Trk B and BDNF involved in the progression of cevical cancer.3, The expression strength of Trk B and BDNF were gradually increased with higher clinical stage and poor differentiated of cervical cancer,which showed that Trk B and BDNF correlated with the malignant behavior of cervical cancer.4,Trk B associated with BDNF in the progress of CINS and cervical cancer,thus they may regulate the progress of CINS and cervical cancer through signal transduction pathway.5,In the process of CINS and cervical cancer, the high expression of Trk B and BDNF both associated with high-risk infection,which indicated high-risk HPV playing a carcinogenic effect probably through Trk B and BDNF play a role in cancer. |
