Objective:The aim of this study is to investigate whether TGF-β1 promotes the expression of HMGA1 through ERK1/2 and or Akt pathway.Methods:In order to elucidate the molecular mechanism whereby TGF-β1 up-regulates HMGA1, we incubated the LX-2 cells with different concentration of TGF-β1,and detected the mRNA level of HMGA1 with Realtime-qPCR.Further,we used TGF-β1and PD98059(MK2206) to treat LX-2 cells,and used western blot to detect the protein level of pERK1/2,pAkt,ERK1/2,Akt and HMGA1, used Realtime-qPCR to detect the mRNA level of HMGA1.Results:1 、 We observed a dose-dependently increase of HMGA1 in the presence of TGF-β1,which was up to a peak at 5ng/ml.2、We showed that TGF-β1 markedly induced the activation of ERK1/2 and Akt pathway.3、We showed that TGF-β1markedly induced the expression of HMGA1,and the inhibitor of ERK1/2 pathway significantly blocked the effects of TGF-β1,while the inhibitor of Akt pathway had no effects.Conclusions:1、TGF-β1 markedly induces the expression of HMGA1 in LX-2 HSC cells.2、TGF-β1 markedly induces the activation of ERK1/2 and Akt pathway in LX-2HSC cells.3、TGF-β1 promotes the expression of HMGA1 through ERK1/2 pathway.
