Purpose:To setup rat lung transplantation model, To defined The expression of Autophagy related protein Beclin-1 in lung ischemia/reperfusion injury after rat lung transplantation, through regulating autophagy, research the change of Beclin- 1protein expression, the apoptosis level and the lung function indices. To explore the influence of autophagy on lung ischemia/reperfusion injury and lung function.Methods:To setup rat lung transplantation model, Rat lung perfusion, cold ischemia and transplantation, after 2h, 6h, 12 h reperfuse respectively and obtain the lung specimens. Detect the level of expression of Beclin-1, apoptosis and the lung function indices. Through regulating autophagy, Use LPD, 3-MA+LPD,Rapamycin+LPD perfuse and preserve the lung respectively, then transplant the lung.At the peak time of autophagy level we got from the first part, reperfuse respectively and obtain the lung specimens. Detect the level of expression of Beclin-1, apoptosis and the lung function indices again.Results:1. Stable pattern of in SD rat lung transplantation was setup. 2. The level of beclin-1 protein expression reach the peak in 6h after lung transplantation, cell apoptosis and lung function injury reach the peak in 6h after lung transplantation. 3.The level of beclin-1 protein expression and cell apoptosis reduce significantly in3-MA groups, reduce lung injury. The level of beclin-1 protein expression and cell apoptosis increase significantly in Rapamycin+LPD groups, increase lung injury.Conclusion:1, The level of autophagy related protein Beclin-1 and cell apoptosis increased,and reach the peak in 6h after lung transplantation.2, By inhibiting the autophagy, The level of beclin-1 protein expression and cell apoptosis reduce significantly, reduce lung injury. The transplant ischemia-reperfu-sion injury has a protective effect by inhibiting the autophagy.3, By activating the autophagy, The level of beclin-1 protein expression and cell apoptosis increase significantly, increase lung injury. The transplant ischemiareperfusion injury has a promotion effect by activating the autophagy.4, Autophagy may has a promotion effect in transplant ischemia-reperfusion injury.
