Objective : For the past years,a large number of studies have shown that polysaccharide of many plants has a variety of physiological functions, but polysaccharides of Nitraria tangutorum Bobr(NTB) and Lycium ruthenicum Murr.(LRM) on T/NK cells is not yet clear. In this study, the dried fruits of NTB and LRM are used as the main raw material, using the ultrasonic-microwave synergistic technology to extract the polysaccharide from NTB and LRM, and then the immunomodulatory effects of polysaccharide of NTB and LRM in immunosuppressed mice, namely, NTB and LRM polysaccharides on the interferon-γ(IFN-γ)and the subset of T lymphocyte, including CD3/CD4/CD8/ and natural killer(NK) cells in mice were studied.Method:1.Using the ultrasonic-microwave synergistic technology to extract the the polysaccharide of NTB and LRM, the final extraction rate of polysaccharide was 47.78% and 35.90 %, respectively. 2. Groups : 40 mice was divided into 4 groups.e.g. control group; model group; model +NTB and model +LRM group. 3. Establishment of immunosuppressed model : CPM was given to establish the immunosuppressed mice model the dose of cyclophosphamide(CPM) was 70 mg/kg, injected it into intraperitoneal for 5days.4.Treatment of the immunosuppressed mice by NTB and LRM: the dose of NTB and LRM were 3.6g/kg continually injected for 10 days.the control group was injected same dose of normal saline. the peripheral blood T cell was detected after 24 h terminal treatment : the weight of spleen and thymus and the amount of CD3/CD4/CD8/NK and serum IFN-γ were detected.5.Defection of murine immune function :(a)The weight of spleen and thymus, the spleen structure were observed;(b)Expression of IFN-γ m RNA was detected by RT-PCR and the concentration of serum IFN-γ was detected by ELISA;(c)The murine peripheral lymphocyte subset including CD3/CD4/CD8 and NK cells were detected by Flow cytometry. Results: compared with the immunosuppressed model group, the weight of spleen and thymus were increased and the structure of them recovered near to normal. Especially, the lymph follicles were increased in NTB and LRM group then those of immunosuppressed model group. The IFN-γ m RNA expression was up-regulated and the serum IFN-γ was significantly increased in the model +NTB and model +LRM group; the amount of CD3/CD4/CD8 and NK cells were remarkably increased then those of immunosuppressed mice. Conclosion: NTB and LRM can obviously increase the weight of spleen, thymus and improve the structure of spleen. Moreover, the NTB and LRM can increase the concentration of IFN-γ and enhance the IFN-γ m RNA expression in immunosuppressed mice.The two kinds of polysaccharides can also increase the amount of CD3/CD4/CD8 and NK cells.
