Changes Of H₂S/Cystathionine Gamma Lyase System Of On The Wound Burned Of Rat

Objective:To study the changes of Macrophages hydrogen sulfide/cystathonine gamma lyase system on the wound of burned rats.Method:78 healthy SD rats were divided into 13 groups(normal control group, blank control group 1,2,3, Na HS group 1,2,3, PPG group 1,2,3, Na HS + PPG group1,2,3); the normal control group received no intervention, the rest of the groups were formed a model of 5% partial thickness burns(confirmed by biopsy), the rats in the blank control group were timely intraperitoneal injected saline every day, the rats in the Na HS group were timely intraperitoneal injected Na HS solution every day, the rats in PPG group were timely intraperitoneal injected propargylglycine daily, the rats in Na HS+PPG group were timely intraperitoneal injected Na HS solution and propargylglycine daily, on the day of 3,7,14 to take macrophage from the burned rats to culture; to measure the contents of hydrgen sulfide in the cell culture supernatant and measure the macrophages CSEm RNA gene expression levels.Results:At the same time point, the cell culture supernatant concentration of hydrogen sulfide, the saline group(NS3, NS7, NS14) and normal control group were significantly different(P<0.05); at the same time point, Na HS group(N3, N7, N14) with the saline group(NS3, NS7, NS14) were significantly different(P<0.05); at the same time point, PPG group(P3, P7, P14) with the saline group(NS3, NS7, NS14) were significantly different(P<0.05); at the same time point, Na HS group(N3, N7, N14) and Na HS + PPG group(N+ P3, N+P7, N+P14) were significantly different(P<0.05); at the same time point, PPG group(P3, P7, P14) and Na HS + PPG group(N+P3, N+P7, N+P14) were not significantly different(P>0.05); at the same time point, CSEm RNA changes during the H2 S secretion process, saline group NS3 compared with the normal control group, there are significant differences(P<0.05), the saline group(NS7, NS14) and normal control group showed no significant difference(P>0.05); at the same time point, Na HS group(N3, N7, N14) and saline group(NS3, NS7, NS14) were significantly different(P<0.05); at the same time point, PPG group(P3, P7, P14) with the saline group(NS3, NS7, NS14) were not significantly different(P>0.05); at the same time point, Na HS group(N3, N7, N14) and Na HS + PPG group(N+P3, N+P7, N+P14) were significantly different(P<0.05); at the same time point, PPG group(P3, P7, P14) and Na HS + PPG group(N+P3, N+P7, N+P14) were significantly different(P<0.05).Conclusion:The function of macrophages hydrogen sulfide / cystathionine Gamma lyase system was reduced on the wound of burned rats. Minimum exogenous hydrogen sulfide has a facilitating role on the macrophages hydrogen sulfide / cystathionine gamma lyase system.PPG can inhibit the formation of endogenous H2 S, but cannot inhibit the CSEm RNA expression...
