| Objectives In this study, all rats were castrated, grouping, and each group of rats gava with black cohosh, sex hormones, and setting up a castration control group with distilled water, to detect the levels of serum sex hormones, detected their uterus, vagina related target organ E2 receptor expression levels, and to explore black cohosh in the treatment of security, and the E2 receptor expression in each rat’s uterus vaginal levels.Methods Thirty Sprague-Dawley female rats aroud 8-week-old, acclimated after two weeks, then all rats of each group were castrated by surgical and removal of the ovaries, all castrated rats restore twenty days(as five estrous cycles), vaginal smears, five days in a row is no longer cyclical changes, determine the success of castration, all rats were randomly divided into three groups(n = 10), as ovariectomized group(control group), estrogen group(E2 group), and black cohosh group(BC group), the rats of three group were treated for: 1 Control group: feed with distilled water, 2 ml, 1/day, 8 weeks; 2 E2 group: rats were fed with aqueous solution of Estradiolvalerate tablets, 2 ml, 1/day, 8 weeks; 3 BC group: gaved aqueous solution of black cohosh, 2 ml, 1/day, 8 weeks. Each group of rats before ovariectomized, the administration of 4 weeks, and administration of 8 weeks, tails were taking blood, extract the serum, determination of estradiol(E2), the follicle stimulating hormone(FSH) and the luteinizing hormone(LH) content by the mean of radioimmunoassay, administration of 8 weeks after the tails blood takend, rats were killed immediately, while the uterine, vagina, immunohistochemistry distribution of its E2 receptor with the method of immunohistochemistry. The experimental datas were statistically analyzed using by the SPSS17.0 software, and then the measurement data expressed as mean ± standard deviation( sx ±). The intergroup differences were analyzed and determined by the method of Variance analysis of repeated measurement, determine group differences between that P <0.05 meaninng statistically significant.Results 1 Before administration, three group of rats serum E2, FSH and LH make controlled, no significant difference(P>0.05). 2 4 weeks after administration: Compared with the control group, the E2 levels of serum of the E2 group were higher(P<0.05),the levels of the serum FSH and serum LH in the serum were lower(P<0.05), the E2 levels of serum of the BC group were no obviously difference(P>0.05), and the low level of serum FSH and LH(P<0.05); And E2 group compared with the BC group, the content of serum E2 about E2 group was obviously higher than that of BC group(P<0.05), but two groups of serum FSH and LH contrast has no obvious difference(P>0.05). 3 8 weeks after administration: Compared with the control group, the E2 levels of serum of the E2 group were higher(P<0.05),the levels of the serum FSH and serum LH in the serum were lower(P<0.05), the E2 levels of serum of the BC group were no obviously difference(P>0.05), and the low level of serum FSH and LH(P<0.05); And E2 group compared with the BC group, the content of serum E2 about E2 group was obviouslyhigher than that of BC group(P<0.05), but two groups of serum FSH and LH contrast has no obvious difference(P>0.05). 4 4 weeks after administration contorled with before administration:the control group around 4 weeks after administration, the levels of serum E2 were obvious lower(P<0.05), the serum FSH and LH were higher(P<0.05); the E2 group around 4 weeks after administration, the levels of serum E2 were higher, the FSH and the LH were lower, but trough by SPSS,the difference were not obviously(P>0.05); the BC group around 4 weeks after administration, the levels of serum E2 were obvious lower(P<0.05), but the serum FSH and LH were no changed obviously(P>0.05). 5 8 weeks after administration, contorled with before administration: the group of control around 8 weeks after administration, the levels of serum E2 were higher, the FSH and the LH were lower,but trough by SPSS, the difference were not obviously(P>0.05); the E2 group around 8 weeks after administration, the levels of serum E2, FSH and LH were no obvious difference(P>0.05); the BC group around 8 weeks after administration, the levels of serum E2 were obvious lower(P<0.05), but the serum FSH and LH were no changed obviously(P>0.05). 6 4 weeks after administration and 8 weeks after administration, the BC groups of serum E2, FSH, LH compared between groups, the above data statistical analysis respectively, there was no statistically significant difference(P>0.05).7 Compared with the control group, the expression of E2 receptor on the uterus and vagina be enhanced(P<0.05); the uterus and the vagina E2 receptor expression on no significant changed(P>0.05); the uterus and the vagina E2 receptor expression was significantly weaker(P<0.05).Conclusions 1 Black cohosh do working in ovariectomized rats showed no estrogen-like effects, does not affect the levels of serum estrogen, but it affects the serum FSH and LH levels; 2 Black cohosh make on the uterus and the vagina, and other related non- estrogen stimulation of target organ-like effect, it does not affect the express of ER on the uterus and the vagina. |
