Polygonum multiflorum thunb is the dry root of polygonum multiflorum thunb which belongs to polygonaceae plants. polygonum multiflorum thunbnot only be used as tonic Chinese traditional medicine in clinic, but also can be used as a diet health care medicine.Stilbene glucoside from polygonum multiflorum thunb is the main efficacy component that makes the hair and beard black, but its effect on tyrosinase activity is divided. And the mechanisms that the impact of Stilbene glucoside on tyrosinase activity is not yet clear. We have to find a simple an effective purification technology of stilbene glucoside, and to explore its effect on tyrosinase activity and the mechanism of action.ObjectiveTo detect the antioxidant activity of multi-component from Polygonum multiflorum Thunb by using the methods of DPPH radical scavenging capacity.To optimize purification technology of stilbene glucoside from polygonum multiflorum thunb.To discuss the effect on activity of tyrosinase of Stilbene glucoside from polygonum multiflorum thunb and its mechanism in action.Methods1.To separate the rough extract of polygonum multiflorum thunb by Using high-speed countercurrent chromatography and then the various components were received. And the DPPH radical scavenging experiments were carried on the various components, finally, antioxidant activity of various components were compared.2.To establish a simple and feasiblepurification technology of high purity of stilbene glucoside by using high-speed countercurrent chromatography.3. The method of rate of dopamine oxidation was used to study the impact of stilbene glucoside on tyrosinase activity.4. To do the enzymatic dynamics analysis for the action of stilbene glucoside and tyrosine, to establish Lineweaver-Burk figure by using the enzymatic kinetic parameters. And then the mechanism of action of stilbene glucoside on tyrosinase was explored.Results1.Four components were received by using HSCCC, which the third component has the maximum activity of DPPH radical scavenging, and was identified as stilbene glucoside, that its purity was more than 95%.2. A simple and feasible high-purity purification technology of Stilbene glucoside was established by alkalifying the crude extract and HSCCC, and the purity of stilbene glucoside can reach 99.15%.3.The result of dopamine oxidation rate showed that the stilbene glucoside in low concentration has the activated effect on tyrosinase(IC50 were0.013 mg/ml, 0.018 mg/ml, 0.007 mg/ml), and in high concentration it has the inhibitory effect on tyrosinase(IC50 were 1.177 mg/ml, 1.046 mg/ml, 1.203 mg/ml).4. The enzymatic kinetic parameters and Lineweaver-Burk figure were made by enzymatic dynamics analysis of tyrosinase. And the mechanism of stilbene glucoside on tyrosinase may be the mixed activation mechanism.ConclusionThe conclusion that the stilbene glucoside was the main component of the antioxidant activity from polygonum multiflorum thunb was received by the DPPH radical scavenging experiments.A simple and feasible high-purity purification technology of stilbene glucoside was established by alkalifying the crude extract and HSCCC.The mechanism of stilbene glucoside impact on tyrosinase activity was discussed preliminarily.
