| Morindae Officinalis Radix as a Lingnan traditional Chinese medicine is the root of Morinda officinalis How. which has a variety of effects such as enriching the kidney and strengthening yang and so on, it also help people to be stronger for those whose feet are not as health as young people,is one of the traditional Chinese medicine which is often used. The researches show that Morindae Officinalis Radix have many different contaminations like anthraquinones, saccharides, amino acid, volatile components, iridoids and iridoid glycosides, steroid chemicals, organic and inorganic etc. The Chinese Pharmacopoeia 2010 Volume I indicats that the contamination of Nstose, moisture and extracta should be tested.Morindae Officinalis Radix are produced in Jiangxi Province, Fujian province, Taiwan, Guangdong province, Hong Kong, Hainan province and Guangxi province, since ancient times, the Morindae Officinalis Radix is the famous-region drug of the deqing country, Guangdong province. As it cited in some records, the Gaoliang town is the most suitable place for planting Morinda officinalis How. There are many limitations of the ways which are used to test tradition Chinese medicine, like consuming so much time, wasting a lot of money, and there are many mistakes made by people and so on. So people are looking for a new method which is faster, more accurate, and do harmless to the environment. The NIRS which satisfying all people’s need is widely used all over the world. Our researching group has the strong background because we had applied this technology in different herbs, such as pericarpium citri reticulatae chachiensis, Pogostemon cablin, folia eriobotryae and the products which are made by them, and established the quantity and quality models. This experiment firstly tested the contaminations of the Nstose, moisture and extraction of Morindae Officinalis Radix. which is directed by the Chinese Pharmacopoeia 2010. And then analyse the connection of contamination and the data of NIRS. We are looking for a method which can determine the quality of the herb fastly, accurately. The first part of this paper under the direction of Chinese Pharmacopoeia2010 Volume I, used HPLC-ELSD to determine the contamination of Nostose in different samples which collected in Wuzhou city, guangxi province, Zhao qing city and yun fu city, Guangdong province. All the samples are qualified. From then on, compare with different areas and different growth periods, we found out that the products grow in zhaoqing city, Guangdong provinces, the contamination is the highest, and those which grow in yun fu city is the lowest. And the best growth period is 4 years. This experiment can tell people the fact that where to grow the herbs is the best, and when is the best to collect them. The second part include three little portions, they are NIR quantitative models of the Morindae Officinalis Radix’s water extraction, moisture and Nystose respectively. At first, we gathered the data of the contamination of the water extraction, moisture and Nystose which were tested by the traditional methods, and then gathered the NIR information of the samples. Later, using the chemometrics like PLS to combine the two data, and picked out the most suitable pretreat methods, and chosed the best wavelength and factors to establish the three quantitative models respectively. The water extraction quantitative model’s R2 is 0.9915, RMSEC and RMSEP are 0.472 and 0.489; the moisture quantitative model’s R2 is 0.9855, RMSEC and RMSEP are 0.169 and 0.180; the Nystose quantitative model’s R2 is 0.9791, RMSEC and RMSEP are 0.900 and 0.909. In order to examine the stability and the availability of the models, we need to evaluate the forecast average recovery rate, precision and repeatability, the three data of the water extraction quantitative model are 99.89%, 0.723%, 1.041%; the moisture model are 100.12%, 1.275% and 1.489%; the three index data of the Nystose model are 100.05%,0.739%,1.866%. So those models which established are stable and accurate and can be put into practical works. |
