Currently, 5- Fluorouracil(5-FU), a typical clinical medication within the fluorouracil family, has been widely used as colon cancer chemo-therapy. However, the therapeutic effect of 5-FU as a single treatment is limited, highlighting the combination of 5-FU and other agents, which could promote better curative effect. Vitamin B, which participates in DNA methylation, neucleotide synthesis, gene repairmen and so forth, is tightly associated with the development and progression of many types of cancer.The present study utilized CT26 transplanted tumor model, aimming at 1) investigating the preventive role of Vb2 in colon cancer treatment; 2) the combination of Vb2 and 5-FU as a post-operation therapy, and how it relates to cancer metastasis; 3) the relationship between tumor operation and colon cancer prognostic. Results show that Vb2 promotes tumor growth in pre-neoplastic mice, which could be explained by two mechanisms including cell metabolism and enhancing ki-67 expression. While after tumor removed by operations, the combined use of Vb2 and 5-FU could significantly reduce ki-67 expression, serum vegf production, and increase E-cad expression.Therefore, it could be concluded that Vb2 treatment in pre-neoplastic organism should not be advovated, since it potentially nourished tunor cells and increase tumor growth. However, in terms of patients with colon cancer, the best treatment should be whole movement of tumor by operation, combined with 5-FU and Vb2 treatment. Since this therapy could decrease the risk of tunor recurrence, improve thymus index, control tumor vessel growth, and inhibit tumor metastasis. The present study could provide data optimizing clinical treatment of colon cancer.
