Objectives Studying on the similarities and differences of Parthenogenetic activation embryos and sperm-egg binding embryonic of mouse in changes of glucose metabolism,and amino acid metabolism, comparing the similarities and differences in process of development of parthenogenetic embryos and sperm-egg binding embryonic,to provide evidence in studing on embryonic development of Parthenogenetic activation of potential.Methods Collect the embryo culture medium on the D1(2-cell stage),D3(8 ~ 12-cell stage),D5(blastula stage),and then useing the glucose oxidase method to determinate the content of glucose in the culture fluid,and using the tandem mass spectrometry method to detect the content of alanine,glutamic acid and L-aspartic acid in the culture fluid,analyse the similarities and differences between parthenogenetic embryos and naturally fertilized embryos in glucose metabolism,alanine metabolism,glutamic acid metabolism and Laspartic acid metabolism.Results 1 The content of glucose in the culture medium on the D1 between parthenogenetic embryos and naturally fertilized embryos( μmol/ml)(-8±0.2;-9±0.2),there is no differences(P>0.05);the absorption of alanine between parthenogenetic embryos and naturally fertilized embryos( μmol/ml)(3±0.2;5±0.2),the absorption of glutamic acid of between parthenogenetic embryos and naturally fertilized embryos(-17±0.2;-19±0.2),the absorption of L-aspartic acid between parthenogenetic embryos and naturally fertilized embryos(-5±0.2;-7±0.2),there is no differences(P>0.05);2 The content of glucose in the culture medium on the D3 between parthenogenetic embryos and naturally fertilized embryos(-23±0.2;-26±0.2),there is no differences(P > 0.05);the absorption of alanine of between parthenogenetic embryos and naturally fertilized embryos(8±0.2;14±0.3),there are differences(P<0.05);the absorption of glutamic acid of between parthenogenetic embryos and naturally fertilized embryos(-39±0.2;-42±0.2),the absorption of L-aspartic acid between parthenogenetic embryos and naturally fertilized embryos(-9±0.2;-11±0.2),no differences(P>0.05);3 The content of glucose in the culture medium on the D5 between parthenogenetic embryos and naturally fertilized embryos(-275±0.2;-584±0.2),there are differences(P < 0.05);the absorption of alanine of between parthenogenetic embryos and naturally fertilized embryos(53±0.2;86±0.2),the absorption of glutamic acid of between parthenogenetic embryos and naturally fertilized embryos(-52±0.2;-94±0.2),there are differences(P<0.05);the absorption of L-aspartic acid between parthenogenetic embryos and naturally fertilized embryos(-23±0.2;-21±0.2),there is no differences(P>0.05).Conclusions 1 The ability of metabolism in glucose. alanine.glutamic acid in mouse parthenogenetic embryos is poor than naturally fertilized embryos on D1-D5 days. 2 The ability of metabolism in L-aspartic acid is poor than naturally fertilized embryos on D1-D5 days.3 Parthenogenetic embryos and naturally fertilized embryos has some similarity,and all of them have development potential.
