Studies On Quality Standard Of Entadae Semen

Entadae Semen(Keteng zi) is the dried seed of Entada phaseoloides(Linn.) Merr. It has the ability of invigorating function, invigorating the stomach and promoting digestion, dispeling wind and relieving pain, strengthening the muscles and bones. It is intended for insufficiency of blood, week, pain, poor digestion. Lots of studies have suggested that Entadae Semen may offer therapeutic benefits such as analgesia antiinflammatory, anti-hyperglycemic, anti-tumor actions. However, the insufficient research of E. phaseoloides can’t control the quality of this crude drug effectively. So it is necessary to establish the quality control method and standard of Entadae Semen, and provide scientific basis for the quality control of Entadae Semen.The components of Phaseoloidin and Entadamide A-β-D-glucopyranoside are the main bioactive compounds in the herb and have the ability of analgesia antiinflammatory and invigorating the blood which have high contents in the plant, so Phaseoloidin and Entadamide A-β-D-glucopyranoside are used as reference standards.By column chromatography, recrystallization, Phaseoloidin and Entadamide A-β-D-glucopyranoside were puried. Through the MS, UV, IR and NMR, their structures were identified. To observe the effect of Phaseoloidin, Entadamide A-β-Dglucopyranoside and ethanolic extract in Qi-deficiency and Blood-deficiency model mice. The result shows that Phaseoloidin, Entadamide A-β-D-glucopyranoside and ethanolic extract have the ability of invigorating function. So the characteristic components from Entadae Semen can make a distinction between the fake stamps and original species.To provide scientific basis for the quality control of Entadae Semen, we establish the quality standard.The methods of Character identification and TLC identification for Entadae Semen were established. After analyzing the 21 batches samples, we can reach a conclusion that the methods are specific for identification of Entadae Semen.The test of water and ash content of Entadae Semen were carried out according to the methods recorded in appendix of Chinese Pharmacopeia(2010 edition, volume 1). According to the examination results of 21 batches samples, the content of water should be less than 9.0%, and the content of total ash should be less than3.0%.The alcohol-soluble extract was detected with cold dipping. According to the examination results of 21 batches samples, the content of alcohol-soluble extract should not be less than 29%.The method of detecting Phaseoloidin and Entadamide A-β-Dglucopyranoside in Entadae Semen by HPLC is established. All of the above data proved that the established standard is acceptable for qulity evaluation of Entadae Semen. The result shows that the content of Phaseoloidin is between 5.12% to 9.24%, he content of Entadamide A-β-D-glucopyranoside is between 0.55% to 2.17%; the limtion of Phaseoloidin and Entadamide A-β-D-glucopyranoside in Entadae Semen should not be less than 4.0% and 0.6%, respectively.
