Tianma Gouteng decoction has been widely used clinically to treat hypertension.Although many researchers in recent years have made the attempt to elucidate the mechanism of it, there is still a long way to make it because of the dialectical treatment of TCM, the complex constitutes of traditional chinese medicine and its characteristics of multi-components, multi-targets and synergistic effects, which is of the biggest obstacle on the way to realize its modernization. With the development of Omics, improvement of computer technology, and accumulation of considerable experimental data, network pharmacology, with systematic and integral characteristics which accords with theory of TCM very well, emerged at such moment. Untill now it has been used widely to study traditional chinese medicine, such as elucidating its mechanism, analyzing the synergism between herbal medicines in a prescription, discovering the new indications, screening potential active components or candidate targets and so on. In this study, we did the research into the Tianma Gouteng decoction treating hypertension by means of constructing and analyzing networks. In the first chapter, we summarzied the development of network pharmacology and process of mechnaism study on Tianma Gouteng decoction in treating hypertension. Additionally, the medication rule of the formulae including uncaria was analyzed through text mining, and the result showed that and its changed form were clinically used most frequently. So we choosen Tianma Gouteng decoction as the object of this study. The system pharmacology tecnology platform and other database sources were then applied to construct the database for Tianma Gouteng decoction. Based on the database established, the components-targets network, targets-diseases network and targets-targets were built successively. After that, network analyzing technologies were used to mine useful information. During the process of analyzing indications of Tianma Gouteng decoction, we got two important targets, COX-2 and ADRB2, and the results indicated the potential application in treating tumor, nerve system diseases, asthma and so on. Furthermore, hypertension disease network was built through extending the knowntargets-seedgene, on the background of HPRD to get more potential targets related to hypertension. Take the usual used herb pairs, tianma-gouteng and gouteng-chuanniuxi as examples and use the network pharmacology technology to analyze the synergistic effect,we finally find that the mechanism of synergistic effect between herbs realized through two main ways:(1) Many constitutes act on the same target;(2) Many constitutes act on different functional targets involved in the same or the related pathway. To elucidate the mechanism of Tianma Gouteng decoction acting on hypertension, 57 intersective targets between Tianma Gouteng decoction and hypertension were analyzed by the ClueGO plugin used in Cytoscape, to make GO and KEGG pathway enrichment analysis. The formula may exert its efficacy by acting on the targets which possess the molecular function such as G-protein coupled amine receptor activity, adrenergic receptor activity, alpha-adrenergic receptor activity, catecholamine binding, epinephrine binding, ammonium ion binding,nitric-oxide synthase regulator activity and so on, playing an important role in Gap junction,cGMP-PKG signaling pathway and Calcium signaling pathway and so forth. |
