| Atrial fibrillation with valvular heart disease(AF-VHD) is a kind of the valvular diseases.Most patients with AF-VHD have severe symptoms because of it will cause a series of diseases easily, such as heart failure, myocardial infarction, thrombus and so on.With the rapid growth of modern molecular biotechnology, the geart progresses in the pathological mechanisms of AF-VHD has been achieved from academic research, but that still be falling short. we need to make comprehensive analysis from integrity of the network in global perspective. Therefore, this paper use the mathematical analysis of network and biological information method to construct two specific co-regulation networks from AF-VHD and valvular heart disease(VHD).Main content is following:( 1) Collecting 15 VHD related miRNAs from papers which has been published,and then we constructed the co-regulation network of VHD based on the strategy of regulation format, which including of mi RNAs,transcription factors(TF) and genes. Four hub miRNAs, four hub TFs and thirty-one hub genes were identified in the VHD regulatory network. First, we analyzed the biological function of 34 hub genes.Then, Using the GO functional enrichment and KEGG database to evaluate the hub miRNAs and target genes. Finally, The results are mapped into biological space to reveal the pathological mechanisms of VHD.(2)Using asymmetric principal componet analysis and Bootstrap method to obtain 32 AF-VHD related mi RNAs from miRNA expression profiles of AF-VHD. We constructed the co-regulation network of AF-VHD also based on strategy of regulation format and identified five hub miRNAs, five hub TFs,forty-one hub genes. We taking similar approaches as VHD regulatory network to show the molecular mechanism of AF-VHD. In addition, We analyzed the commom biological features in both VHD and AF-VHD networks and mined the biological information of diseases to build pathological mechanisms from VHD to AF-VHD. |
