Carboxymethyl chitosan is a water-soluble derivative of chitosan. Because of its a varietyof biological activities such as anti-virus eliminate inflammation etc. It has been widely used. carboxymethyl chitosan Zn is a water insoluble carboxymethyl chitosan metal complex. Its structure and excellent antibacterial effect has been reported. Because Carboxymethyl chitosan can accelerate the wound healing and zinc ions is also essential for wound healing, carboxymethyl chitosan Zn(11) is expected to be a kind of wound dressing with skin repairing function. However, there is few literature on this subject. In this paper, biological safety and functionality of carboxymethyl chitosan Zn complex were investigated to provide a basis for a new type of biological materials for wound healing.Carboxymethyl chitosan and zinc chloride were used as the raw material to get carboxymethyl chitosan Zn complex material. Biological safety was evaluated by intracutaneous stimulition test in rabbits, cytotoxicity test using human fibroblasts, subacute toxicity test by intraperitoneal injection. In vivo study, rat scald burns model and rat model of chronic diabetic ulcers were established to observe therapeutic action of the material. Wound of scald burns model was third-degree burns. Hydroxyproline, TGF-β and IL-6 content in tissue were measured. Wound of model of chronic diabetic ulcers was full-thickness cutaneous wounds created by incision. Hydroxyproline, VEGF and CD34 content in tissue were measured.Results reveal that the material has favorable safety. Ⅲ degree burn wound healing experimental results show that the carboxymethyl chitosan zinc complex material can promote synthesis of hydroxyproline and collagen fibers deposition at early phase of healing and reduce the wound scar at the end of healing by regulate the secretion of TGF-βand IL-6 effectively. Cchronic diabetic ulcers healing study results show that carboxymethyl chitosan zinc complex material can increase the healing wound microvascular content and accelerate the collagen deposition by increasing the content of VEGF and CD34 in the wound tissue.In conclusion, carboxymethyl chitosan zinc complex material has good safety. The material can create a relatively closed and humid environment for wounds, besides, it can shorten the healing time of acuteand chronic wound by regulating cytokines secretion, Meeting the requirements of new biological wound dressing.
