Background and objective:Nowadays the society of human cardiovascular disease incidence increased, mortality is also rising, heart disease is the most common fatal diseases, because of its strong burst so great harm, is one of the leading causes of human death. Aiming at the complex congenital heart diseaseand serious heart disease, cardiopulmonary bypass operation is currently the most effective therapy, and cardiopulmonary by pass technique appears to provide a firm foundation for the heart stopped jump in open heart operation; in cardiac arrest under open heart operation, the most critical is to protect on myocardium. Since the 50’s of last century, cardioplegia began to be heart surgeons use in cardiac surgical operation, cardioplegia can induce heart in temporary arrest, mechanical action to stop making the heart patients can get more clear operation visual field during operation, more conducive to the surgery operation.Cardiac arrest and resuscitation is bound to cause myocardial is myocardial ischemia reperfusion injury, myocardial protection due to improper operation after the first heart disease mortality, myocardial protection is an important prerequisite for so successful heart operation. The 70 years since the last century, clinicians began using cardioplegia myocardial protection function, which makes the complex open heart operation can be carried out smoothly. In recent years more and more scholars cardioplegia and myocardial ischemia / reperfusion injury(MIRI) of interest, after several decades of research, all kinds of cardioplegia and arrest methods appear continuously, cardioplegia during cardiac surgery operation has also been widely used, become important protection measure of myocardial protection, at present already in-depth study of cardiac arrest andreperfusion stroke liquid formulation technology,but which method best effects remain controversial.The purpose of this experiment was to study the effect of temperature and potassium ion concentration on coronary artery smooth muscle,anatomy and physiology of the human body to choose is very similar to the pigs as experimental object, through the contrast of pig coronary artery smooth muscle at a certain temperature for different concentrations of potassium ion reaction, to provide experimental reference for various types of cardiac arrest in the application of cardioplegia clinical on.Materials and methods:Select healthy Swedish 24 pigs, male or female, weight 46 + 3kg. Each experiment selected 2 head, were obtained from the anterior descending branch of the coronary artery vessels, divided into 1mm long pieces, each pig randomly selected from each of 3 sections, respectively, for organ bath experiments(organ bath). Adjust(bath temperature were set as 4 DEG C, 20 C, 30 C, 37 C),until the bath temperature is constant, added to the same temperature of high potassium solution Krebs(K concentration 127mmol/L) stimulates vascular contraction for two times, and record the maximum systolic coronary vascular smooth muscle(Cmax), Krebs value of solution adding different potassium ion concentration to the bath in the(K concentrations were 8, 12, 16, 20, 24, 32, 40,48, 56, 64mmol/L), record the maximum tension of each concentration induced value respectively, and the maximum shrinkage(Cmax) contrast, calculated for each concentration induced coronary artery shrinkage shrinkage(Cmax)accounted for the largest percentage of each sample, self control, determination of four coronary artery induced by different concentration of potassium ion temperature shrinkage degree.Results:1 With the increase of temperature, coronary vascular smooth muscle of potassium induced contraction of more sensitive;2 When the temperature is 37 degrees centigrade, potassium ion concentration in the following 12mmol/L induced coronary vasoconstriction is not obvious;3 When the temperature is 30 degrees centigrade, potassium ion concentration in the following 16mmol/L induced coronary vasoconstriction is not obvious;4 When the temperature is 20 degrees centigrade, potassium ion concentration in the following 24mmol/L induced coronary vasoconstriction is not obvious;5 When the temperature is 4 degrees centigrade, potassium ion concentration in the following 64mmol/L induced coronary vasoconstriction is not obvious;Conclusion:1 open heart cardiac arrest operation, can make the heart arrest in smoothly on the basis of the potassium ion concentration is lower better heart perfusion;2 the use of warm blood cardioplegia perfusion method in cardiac arrest cardioplegia potassium ion concentration should not be too high;3 Cold crystalloid cardioplegia hypothermic perfusion does not cause coronary vasoconstriction of beating heart, can get satisfactory perfusion effect.
