Objective:(1) To understand the control effect of extract liquid of glucocorticoid induced osteoporosis prevention effect of Guilu Erxian, comparing alendronate sodium, and the effects on bone metabolism of rats, to understand the effect of extracted liquid on serum biochemical indexes of Guilu Erxian glue, as well as on osteoblast and osteoclast function.(2) and to detect its effect on bone density in rats, to analyze its main mechanism of action.Methods: From the experimental animal center of Zhengzhou University selected 80 3.5 month old SPF female SD rats(200 ± 20g), were randomly divided into A, B, C, D4 group, 20 rats in each group, the control group of A group, the model group of B group, Chinese medicine group was C group, western medicine group for the D group; group B, group C, group D were intravenously injected with Dexamethasone Sodium phosphate Injection(2.5mg / kg.d, 2 times / week), Dexamethasone Sodium phosphate Injection(2.5mg / kg.d, 2 times / week) + extract Guilu Erxian(3.5ml /kg.d) by gavage, Dexamethasone Sodium phosphate Injection(2.5mg / kg.d, 2 times / week) + alendronate sodium(1mg/kg.d) by gavage, group A, group B with normal saline at the same dose by gavage, medication period of 8 weeks. After 8 weeks of concentration of chloral hydrate intraperitoneal injection of 8% after anesthesia, from abdominal aortic puncture to draw blood, centrifugal evacuation of laboratory testing, tartrate resistant acid phosphatase(TRACP-5b), serum bone alkaline phosphatase(BALP), molecular fragments(N-MID- OT end), hydroxyl(25- Vitamin D3(25 OH) D3) content, take the left femur and lumbar vertebrae in rats five, removal of the surrounding soft tissue, bone density of lumbar and femur of rats in each group(BMD) value. Data obtainedafter the application of statistical analysis software analysis.Results: The confirmed by application of glucocorticoids and after drug treatment, B group spirit and hair color gloss than other groups of rats was significantly lower than that of other groups, the difference of body weight, the situation of C group before treatment than that before improvement, the D group than before modeling improved slightly but less than C group obviously; the detection results: B group TRACP-5b, the concentration of N-MID-OT was significantly higher than other groups(P, <0.01) BALP and 25(OH) D3 concentrations were significantly lower than those in the other groups, D group of TRACP-5b, the concentration of N-MID-OT was higher than A group and C group(P >0.05), D group, BALP 25(OH) D3) and the concentration is lower than A group and C group.(P <0.01), C treatment group were significantly better than D group and B group(P, <0.01) and group A(P <0.01) is most close to; bone density: C group was significantly stronger than that of B group(P <0.01), in D group than C group and A group(P <0.01), but the strong in the B group(P <0.01);Conclusion: Guilu Erxian extraction liquid can improve the activity of osteoblasts, promoting bone formation, inhibit osteoclast activity, inhibit bone resorption, improving calcium, phosphorus metabolism, thus weakening of glucocorticoid on bone formation, inhibition of osteoclast role, to achieve the prevention and treatment of GIOP.
