Objective: to research the expression levels of the human telomerase reverse transcriptase TERT and the human protection of telomeres TIN2 and POT1 in the bone marrow mononuclear cells of the patients with non-severe aplastic anemia. To discuss the relationship between them and then to research the connection with the syndrome differentiation of TCM so as to provide scientific basis on the development, treatment and prognosis with aplastic anemia exactly.Methods: to collect the bone marrow mononuclear cells of 20 patients with non severe aplastic anemia who met with the inclusion criteria(8 cases with kidney-yang deficiency, 6 with kidney-yin deficiency and 4 with kidney yin-yang deficiency) and to extract the RNA. To collect the cells of another 10 healthy people as the normal control group. Using real-time fluorescence quantitative polymerase chain reaction(real-time quantitative polymerase chain reaction, RT-qPCR) to detect expressions of the human telomerase reverse transcriptase TERT and the human protection of telomeres TIN2, POT1 as well as the mRNA of the control group. Then to analye the relationship with the syndrome differentiation of TCM.Results: 1. Compare the the patients of non-severe aplastic anemia with the control group, the expression level of TERT mRNA rises obviously showing a significant difference(P<0.05); the expression level of both TIN2 mRNA and POT1 mRNA reduce evidently which has a significant difference(P<0.05).2. Compared with the control group, the gene expression level of TERT of patients with kidney-yang deficiency, kidney-yin deficiency and kidney yin-yang deficiency rises obviously(P<0.05); while the level of patients with kidney-yang deficiency rises higher than the other ones. The expression level of POT1 and TIN2 with kidney-yin deficiency and kidney yin-yang deficiency changes lower than control group(P<0.05). However, the level of kidney-yang deficiency rises higher than another two(P<0.05).Conclusion: 1. The expression level of ERT mRNA increases more significantly than the control group, while the level of TIN2 and POT1 mRNA reduces. It is possible that TERT、TIN2 and POT1 participate the pathogenesis of non-severe aplastic anemia. 2. The expression level of TIN2, POT1 and TERT mRNA shows lower in the kidney-yin deficiency and kidney yin-yang deficiency syndrome, while the kidney-yang deficiency shows the highest. It suggests that the patients with kidney-yin deficiency and kidney yin-yang deficiency can have more serious lesion than the other one. That is to say, the wastage degree of telomere and apoptosis of hematopoietic cell are likely to be more serious.
