Gastric cancer is a common malignant tumor in our country, which causes serious damage to human health, its development involves many factors and is a multi-stage process.Helicobacter Pylori(Hp) infection is recognized as an original factor in the development of gastric cancer. Hp After it infects gastric mucosal tissue, it can cause a series of abnormal expression of gene and protein by inflammatory reaction and release toxins medium, thus further promote the development of the occurrence of gastric cancer.Accordingly, in this paper, by testing the expression of β- catenin,SOX2 and Cyclin D1 protein in different stages of gastric mucosa tissue to analysis and investigate the relationship between them, aims to further explore the pathogenic mechanism of Hp infection.AIM: To test the expression of β-catenin, SOX2, and Cyclin D1 protein in different pathological changes of gastric mucosa tissue, and to explore the correlation of the them and their relationship with Hp infection. Methods:Paraffin-embedded tissue samples were retrospectively collected from 105 patients,which included chronic non atrophic gastritis group(the NAG,group 1), 35 cases of intestinal metaplasia(IM, group 2) and 35 patients with gastric carcinoma(GC,group 3), who received gastric biopsy and resection specimens at The First Affiliated Hospital of Hebei North University.Hp infection was detected by 13C-urea breath and Giemsa. The expression of β catenin, SOX2 and Cyclin D1 protein in the above mentioned tissues were immunhistochemically examined. Results: 1. The infection rate of Hp in the NAG, IM and GC group were 28.5%, 54.3% and 85.7%, showed a trend of increase, there was significant difference between groups(P < 0.05). 2. The expression rate of β-catenin protein in the tissues of NAG, IM and GC were 34.3%, 82.9%and 11.4%, the expression in GC was obviously higher than that in the NAG and IM, there was significant difference between groups(P < 0.05). 3. The expression rate of SOX2 protein in the tissues of NAG, IM and GC was 85.7%,57.1% and 20.0%, the expression of SOX2 protein in GC was lower than that in NAG and IM, there was significant difference between groups(P <0.05). 4. The expression rate of Cyclin D1 protein in the tissues of NAG,IM and GC was 0%, 31.4% and 88.6%, the expression of Cyclin D1 protein in GC organization was significantly higher than that in NAG and IM, there was significant difference between groups(P < 0.05). 5.In gastric cancer,Hp infection and the expression of β catenin and Cyclin D1 was positive related, wihile negative related with the expression of SOX2(P<0.05);SOX2 was found negatively correlated with the expression of β catenin and Cyclin D1 protein(P<0.05); and the expression of β-catenin was positive related with the expression of Cyclin D1(P < 0.05). Conclusion: 1. The infection rate of Hp in IM and GC organization increased significantly than that in NAG group. In the process of NAG- IM – GC, Hp infection plays an important role, and is one of the risk factors of gastric cancer. 2. In NAG,IM and GC, the expression of β-catenin and Cyclin D1 protein were gradually increased, while the expression of SOX2 protein was gradually reduce, they involved in the occurrence of the development of gastric cancer and have a certain correlation. 3. Hp infection, β-catenin, SOX2 and Cyclin D1 proteins are involved in the occurrence and of development gastric cancer, they have their own mechanism of action and synergy jointly to promote the occurrence of gastric cancer.
