Given current DNA methylation in gastric cancer are more concentrated in a single or a few genes, and it is not clear about DNA methylation invasion and metastasis of development in gastric cancer.Therefore, There are great significance to use high-throughput methylation chip and explore their relations in gastric histological type,tumor location, depth of invasion, lymph node and peritoneal metastasis and prognosis. GO annotation classification and KEGG PATHYWAY annotation analysis are able to establish a DNA methylation profiles of gastric cancer and explore gastric cancer exploration, development of epigenetic mechanisms. They intend to establish a molecular staging and prognosis of gastric cancer molecular marker group, so that the target molecule design interventions for the treatment of gastric cancer has opened a new path.Analyzing the methylation status of gastric cancer and their matched adjacent normal tissues use methylated DNA combined NibleGen chip.Using software analyze gens by GO and KEGG PATHWAY annotation analysis.Located in the gene promoter regions, differentially methylated genes presenting in gastric carcinoma are 113, such as SHP1,FGF8 and CSF2 RA.Differentially methylated genes presenting in their matched adjacent normal tissues are 161, such as TNF, IGF2 and BMP7. Located in the CpG islands, differentially methylated genes presenting in the gastric carcinoma are 123, such as WNT2 B, JAK2 and TPT1. Differentially methylated genes presenting in their matched adjacent normal tissues are139, such as TNFRSF4, HOXC8 and NFYA. These genes are located on the different chromosomes. In gastric carcinoma, the state of genemethylation differences at most on the 1st and the 4th chromosomes, both are 11. The state of gene methylation differences at least on the 18 th and on the 20 th chromosomes, both are 1. In their matched adjacent normal tissues, the state of gene methylation differences at most on the11 th is17.The 9th, 19 th and 20 th chromosomes, both are 11. The state of gene methylation differences is 0 on Y chromosome.Many genes are classified the same genes according to function by GO(Gene Ontology) analysis software. There are many fuctions, the protein phosphorylation related Gene, regulate cell decomposition metabolism related genes, the ion transport related gene, enzyme activity regulating genes, the transcriptional regulation of genes, metabolism related genes, cell division related gene,the cell cycle regulation of genes, signal transduction related genes and so on. Pathway gene analysis showed that DNA methylation differences mainly involved in the occurrence of tumors, apoptosis, immunity, amino acid metabolism and so on.Pathway genetic analysis showed that DNA methylation differences mainly involved in the occurrence of tumors, for example: Pathways in cancer, JAK- STAT signaling pathway, Apoptosis, Oocyte meiosis,Tryptophan metabolis, Calcium signaling pathway, Chemokine signaling pathway, TGF-beta signaling pathway, Fc gamma R-mediated phagocytosis.There are significant differences between gastric cancer tissues and their matched adjacent tissues in DNA methylation. DNA methylation locates on different chromosomes, and their numbers and distribution vary widely. The abnormal expression of these genes may be involved in the development of gastric cancer. Genomic DNA methylation differences genes associated with the occurrence and development of gastric cancer.
