| Obesity increases the morbidity and mortality of type 2 diabetes, hypertension, fatty liver disease, stroke, cancer and the other diseases, seriously harmful to human health, has become a major public health problem. Smilax china L.(SCL) is a common traditional Chinese medicine, has the effects of antioxidation, antitumor, antibacterial, anti-inflammatory and analgesic. At present, there is little study about SCL and obesity, this experiment was to study the antiobesity effect of Smilax china L. ethanol extract(SCLE).The SCLE for animal experiment was preparation and analyzed. 50 female ICR mice were randomly divided into 5 groups, fed on normal diet,high-fat diet and high-fat diet plus low, middle, high dose of SCLE for 8 weeks, respectively. Body weight and food intake were determined during this period. Organs and IPAT were weighed at the end of feeding period. Serum lipids concentrations(TC, HDL-C, LDL-C, TG), liver lipids content(TC, TG) and the activities of lipid metabolism related enzyme(FAS, ACC, CAT, ACO, AMPK)were determined. The lipid metabolism related gene(FAS, ACCα, HMG-COAR, HSL, ATGL, CAT, ACO, AMPK, PPARγ2, C/EBPα, SREBP1c) and protein expression(FAS, HSL, P-HSL, ATGL, AMPK, P-AMPK) of in liver and IPAT were determined. The results were as follows:(1) The contents of total triterpenoid, total polyphenol and total flavonoids in SCLE are 14.03±0.21, 42.54±0.45 and 29.69±0.37%. The contents of chlorogenic acid, astilbin, engeletin and resveratrol are 13.65±0.19, 23.65±0.36, 7.97±0.03 and 7.46±0.15 mg/g.(2) SCLE reduced remarkably the body weight gain and IPAT weight in mice.(3) SCLE significantly decreased the contents of TC, LDL-C and TG, increased the content of HDL-C in serum; reduced the contents of TC and TG in liver.(4) SCLE significantly increased the activities of CAT, ACO and AMPK, and did not affect the activities of FAS and ACC in liver.(5) SCLE caused an upregulation of the HSL, ATGL, CAT, ACO and AMPK genes expression, significantly downregulation HMG-COAR and SREBP1 c, and did not affect the expression of FASã€ACCαã€PPARγ2 and C/EBPα in liver and IPAT.(6) SCLE caused an upregulation of the HSL, P-HSL, ATGL, AMPK and P-AMPK protein expression, and did not affect the expression of FAS in liver and IPAT.SCLE mainly through regulating the activities gene and protein expressions of lipid metabolism-related enzymes in liver and adipose tissue, reduced the cholesterol synthesis, promoted fat oxidation, reduced the lipids level in serum and liver, suppressed the fat accumulation and body weight gain. These results suggested SCLE has potential prevent obesity and lifestyle-reated disease. |
