[Objective] culture PC12 cells under different temperature, detect the expression of glutamate receptors in protein, gene level in PC 12 cells under different temperature and illustrate the mechanisms of deep hypothermic brain protection.[Method] randomly divided pc12 cells into four groups: ①deep hypothermia group (culture cells under 20 ℃,5%C02 culture for 3 days), ②moderate hypothermia group:(culture cells under26 ℃,5%C02 culture for 3 days),③ mild hypothermia group (culture cells under 30 ℃, 5%C02 culture for 3 days)). ④The normal temperature group (culture cells under 37 ℃, 5%C02 culture for 3 days)). Each group include 12 samples (6 samples with Western-Blot,6 samples with RT-PCR). Using Western blot technique and real-time PCR to detect the expression of mRNA and proteins of GluR2 and mGluRl-a in different temperatures,the results were analyzed by SPSS 17.0[Results] (1) PC 12 cells can grow well with outside temperature is 20-37℃. (2) The expression of glutamate receptor2 (GluR2) in gene and protein levels of hypothermia group were higher than the amount of normal temperature group and the lower temperature the higher expression, the difference was statistically significant (P< 0.05). (3) The expression of metabotropic glutamate receptor (mGluRl-a) in gene and protein levels of hypothermia group were lower than the amount of normal temperature group and the lower temperature the lower expression, the difference was statistically significant (P< 0.05).[Conclusion]:(1)This model can be easily repeated and can simulate the process of low temperature treatment (2) the temperature can influence the expression of glutamate receptor. (3) low temperature increase the expression of glutamate receptor 2(GluR2)protein formation and with the lower temperature the expression of glutamate receptor 2 (GluR2) in gene and protein level increased gradually, (4) low temperature inhibited mGluRl-a receptor protein formation and with the higher temperature the expression of metabotropic glutamate receptorl-a(mGluRl-a) in gene and protein level decreased, (5) low temperature increase the expression of GluR2 and decrease the expression of mGluRl-a, this may be one of the mechanisms of deep hypothermia protection.
