| ObjectiveThis paper aims to explore the gene polymorphisms on GRIN2A (rs 1070487, rs4587976, rs1650420), OPRD1 (rs581111, rs2236857, rs2236855), CREB (rs3025684) and HTRIB (rs130058) in the male heroin dependence of Yunnan Dai population and their correlationMethodsAccordance with the Diagnostic & Statistic Manuel of Mental Disorders â…£,DSM-â…£.106 heroin dependence males of Yunnan Dai population from Xishuangbanna labor camp were selected as the heroin dependence group, correspondingly 100 healthy males of Yunnan Dai population were selected as the control group. The genomic DNA of the individuals were extracted by using the phenol-chloroform method. A 8 SNPs co-amplification panel was established, and adopting the SNaPshot platform detected the genotype and allele frequencies of the 8 loci candidates on GRIN2A, OPRD1, CREB and HTRIB genes for both the heroin dependence group and the control group. Statistical analysis was processed based on SPSS 17.0, Haploview4.2 and SHESIS software bags.Results1. The result of single locus analysisrs1070487ã€rs4587976ã€rs2236857ã€rs2236855 were detected one genotype, another 4 SNPs were detected three genotypes in heroin dependence group and the control group. There was significant genotype frequency difference of rs581111, rs3025684 and rs130058 between the study group and control group (P<0.025); There was allele frequencies significant difference of rs3025684 between the study group and control group (P<0.05). The allele A frequency of rs3025684 in study group was significantly higher, and the genotype GG frequency of rs3025684 in study group was significantly lower. There were no significant genotype and allele frequencies difference of rs1070487, rs4587976,rs1650420, rs2236855, rs2236857 between the study group and control group.2. The result of linkage disequilibrium analysisIn males of Yunnan Dai population, there was weak linkage disequilibrium between 2 SNPs of chromosome 16 in heroin dependence group and the control group (D’<0.8)3. The result of haplotype analysisThrough haplotype analysis of 2 SNPs of chromosome 16 in the males of Yunnan Dai population, there was significant haplotype frequency distribution difference between the two groups in haplotype of A-Aby rs1650420-rs3025684 (P<0.05). The frequency of haplotype A-A in study group was significantly higher than the control group.ConclusionsThe polymorphisms of GRIN2A (rs581111) and CREB (rs3025684)and HTR1B (rs130058) may be associated with heroin dependence in Yunnan Dai population. And the genotype GG of rs3025684 may be a protective factor of heroin dependence in Yunnan Dai population, the individual with A allele of rs3025684 may be easier to produce heroin dependence. The haplotype of A-A by rs1650420-rs3025684 may be a risk factor of heroin dependence. rs1070487〠rs4587976 polymorphism of GRIN2A gene locus and rs2236857ã€rs2236855 polymorphism of OPRD1 gene locus may be have specificity in Yunnan Dai ethnic group. |
