Background: Irritable bowel syndrome( IBS) is a disorder of gastrointestinal function disease, accompanying long-term abdominal pain、distension and the changing in bowel habit, in addition,it only can be diagnosed after excluding bowel disorders. There is no specific anthropological modification or cardiometabolical abnormalities which can be performed to diagnose irritable bowel syndrome,therefore there are inherent difficult indiagnosing IBS.Meanwhile,IBS is one of high morbidity rate disease and regularly in creased. Base on the statistic,morbidity rate of IBS patients in the western countries is reached at 10%-20% and in our country is also up to 10%. Because of this disease, human beings is decrease their life qualities. Furthermore,the morbidity rate of the youth is higher seniors’ and the morbidity rate of women is twice higher than the men. IBS is complicate and vario,it will lead to mental pressue. In fact that more and more people are pay attention to IBS effects in their life. Subsequently, their clinical and exp erimental research is developing in order to finding the exactly the reason of IBS and make a effective treatment. The reason of IBS is undefined,alternatively,it is may effec ted by disorders、mental、visceral anomalies、brain gut axis and genetic factors.Objective: To analyze the diversity of intestinal flora was carried out in patients with IBS and healthy people affected by IBS. Moreover,compared and contracte IBS patients with healthy people as result of differences of total white blood cells 、 lymphocytes percentage、monocytes percentage、granulocyte percentage.This will be discussed the specific changing of IBS patients immune cell index.Methods: 1. Case choice: according to the Roman Ⅲ diagnostic criteria to determine the cases of IBS, and collect the excrement of IBS patients with IBS. The 24 cases of IBS come from dalian zhongshan hospital affiliated to dalian university,who were in the digestive department ward in hospital from April 2014- September 2014, they aged 21 to 70 years old and 10 cases from men, 14 cases from women. 2. Excrement samples collection: all specimens taken from the patients of IBS early in the morning,and the samples need to make sure that no pollution and natural defecation,we need to pick about 5g excrement samples to the boxes which in ethylene oxide sterilized, sealed specimens placed in frozen- 80 ℃ immediately. The time of acquisition process strictly controlled within 30 min, to prepare for the analysis of intestinal flora. 3. Blood samples collection: early in the morning, using the method of vacuum blood collection to phlebotomize the patients of IBS that keep the fasting state, transfused venous blood 2 ml into EDTA- K2 anticoagulant tube(explained to the patients the time to begin fasting depending on the time from ten o’clock in the evening to the next morning,and they should avoid strenuous activity),blending gently,then taken the samples to laboratory medicine immediately, to clinical for routine blood test(examination of WBC、LY%、MO% and Neu%),the type of machine is Sysmex XE- 2100 automatic blood detector. 4. Comparison and contrast of total white blood cells、lymphocytes percentage monocytes percentage、granulocyte percentage from irritable bowel syndrome patients with the reference values. 5. Using PCR- DGGE fingerprint technique to analyze the differences of intestinal flora between IBS patients and healthy people. 6.Statistical methods: by using the software with SPSS19.0、Quantity One 4.6.2、Image Lab、Canoco 4.5 and so on.Results: 1. The variation of immune cells compare with normal group data,the data of IBS groups’ WBC、LY%、MO% is decreased significantly by P<0.05 except Neu% is ineffective statistic.2. The variation of intestinal bacterial flora compare with health people,the electrophoretic pattern bands of IBS patients is 19.00±2.83, the electrophoretic pattern bands of healthy people is 26.79±3.46,IBS patients DGGE electrophoretic pattern bands are declined by P<0.05 dramatically. PCAanalysis show that normal group and IBS group are slightly difference.Conclusions: 1.These results suggest that IBS patients had lower immune cells、WBC、LY%、MO%than healthy people. 2.IBS patients have intestinal dysbacteriosis and the reduction of the intestinal flora biodiversity. 3.IBS parients’ level of immune cells of the intestinal flora dicersity,the diminution may affect their immune funcions, leading to the onset of IBS.
