Objection:Through digestion of human pancreas from cardiac death donor, we look forward to get enough good quality islets for islet purification, clinical islet transplantation, even the treatment of diabetes.Methods:Our professional organ procurement team gets cardiac death donor pancreas, and keeps in the4℃organ preservation solution (UW solution or HTK solution). Then the pancreas will be deliveried to our laboratory, our team will remove useless pancreatic tissues, exposed the pancreatic duct. Then we will infusion pancreatic digestive juices in the pancreas for10minutes. Based on the collagenase with semi-automatic pancreatic digest system, we will obtain islet cells. After the DTZ dyeing, we count islets number and use it for islet purification. Results:Our team have gotten10pancreas from DCD donors with proper perfusion. We isolated all10pancreases and counted islet number8pancreases. And we obtained an average of712068±712068islets, average islet equivalents for635458±510168IEQ.Conclusion:Through the experiment, we established a reliable set of pancreatic digestive system to get human islets for islet purification, function identification, and laid a foundation for further islet transplantation.
