| Objective Coptis paste treat the folliculitis patients, compare to who are treated with mupirocin ointment. The Coptis paste effective constituent berberine hydrochloride act on the resistant Staphylococcus aureus,through different means to observe the effect of berberine hydrochloride on the status of bacteria and biofilm morphology. Above all, this project may offers a evidence for the berberine hydrochloride’s medicinal value,also offer a new idea for the treatment of resistant Staphylococcus aureus.Methods 1.Choose 60 folliculitis patients, 30 patients were treated with coptis paste and 30 patients were treated with mupirocin ointment,evaluation of curative effect.2.We have selected 4 stains resistance Staphylococcus arueus in Wuhan No.1 Hospital from January to June in 2014, and purchased 1 standard strains of Staphylococcus aureus, ATCC25923,from American type culture collection(ATCC). The berberine hydrochloride and 5 strains of Staphylococcus aureus were co-incubated for 24 hours,and determinated the minimal inhibitory concentration(MIC) values.3. The suspended state of each 5 stains was co-incubated with the concentration of 2 MIC, MIC and 1/2 MIC of the berberine hydrochloride, respectively, after which the growth curve was drawn.4. Similarily, each of biofilm state of 5 stains was co-incubated with the concentration of MIC, 1/2 MIC, 1/4 MIC and1/8 MIC, respectively, after which bar charts were drawn by crystal violet staining and multifunctional enzyme mark instrument readings.5. The suspended state of each 5 stains was co-incubated with the MIC concentration of the berberine hydrochloride, then we observe the plates by transmission electron microscope.6. At the same time, each biofilm state of the 5 stains was co-incubated with the MIC concentration of berberine hydrochloride,after that we observe the plates by scanning electron microscope.Results 1.The patients treated with the coptis paste show the similar effect with mupirocin ointment.2.The berberine hydrochloride can effectively inhibit the suspended state and formation of biofilm of the resistant Staphylococcus aureus in MIC concentration. As the concentration decrease,the inhibit effect also decrease.3.The adhesion of Staphylococcus aureus was disrupted by the MIC concentration of berberine hydrochloride, which effectively inhibits the formation of biofilm. As the concentration decrease,the inhibit effect also decrease.4. Under the transmission electron microscopy, we found the destructive effect of the berberine hydrochloride on the micro morphology of resistant Staphylococcus aureus.5. Under the scanning electron microscopy, we found the destructive effect of the berberine hydrochloride on the micro morphology of resistant Staphylococcus aureus.Conclusion The coptis paste has a great treatment effect on the folliculitis patients.In our research, the suspended state and formation of biofilm of the resistant Staphylococcus aureus were influenced by berberine hydrochloride, which destroyed the normal structure and adhesion between the bacteria. Berberine hydrochloride, effective component extracted from Chinese medicine,is a worthy of clinical application, showing the indicating function for the clinical treatment of the resistant Staphylococcus. |
