Study On Effect Of Yiaikang Capsule On HIV Infected People In The Peripheral Blood Of The Patients With Th22 Cells And The Expression Of Key Transcription Factor AhR

Objection 1 Analyse the changes of Th22, Th1, and Th2 cells in the peripheral blood from HIV infected people, and the relationship between helper T cell subsets and HIV infected people.2 Analyse the expression of Aryl Hydrocarbon Receptors(Ah R),which is Th22 cell transcription factor,in the peripheral blood of HIV infected people, in order to explore the effect of Yiaikang capsule to the expression of Ah R.Methods Screening 30 cases that HIV antibody confirmatory test positive from AIDS Research Center of the First Affiliated Hospital of Henan University of Traditional Chinese Medicine, give the treatment of Yiaikang capsule for a period of 6 months. Draw the peripheral blood from HIV infected people before and after 6 months treatment, and detect the CD4+T lymphocytes. Detect the percentage of Th1(CD4+IFN-γ+), Th2(CD4+IL4+) and Th22(CD4+IL22+) cell in peripheral blood by flow cytometry(FCM). At the same time, detect the relative expression of transcription factors T-bet m RNA, GATA3 m RNA and Ah R m RNA in the peripheral blood mononuclear cells(PBMC) by RT-PCR technology.Results 1 CD4+T lymphocyte: The CD4+T lymphocyte count increased slightly after the treatment of Yiaikang capsule, but has not yet reached statistical significance(P>0.05).2 Th1(CD4+IFN-γ+) cells: The percentage of Th1 cells accounted for CD4+T cells increased after treatment of Yiaikang capsule, with statistical significance(P<0.05).3 Th2(CD4+IL4+) cells: The percentage level of Th2 cells accounted for CD4+T cell after treatment of Yiaikang capsule was not statistically significant compared with before treatment(P>0.05).4 Th22(CD4+IL22+) cells: The percentage of Th22 cells accounted for CD4+T cells increased after treatment of Yiaikang capsule, but with not statistical significance compared with before treatment(P>0.05).5 Expression of transcription factors: The expression of T-bet m RNA increased after treatment compared with before treatment,with statistical significance(P<0.05); GATA3m RNA was not statistically significant(P>0.05); Ah R m RNA increased, was not statistically significant(P>0.05).Conclusion 1 Yiaikang capsule has the potential advantages of improving the percentage of Th1 cells accounted for CD4+T cells and the expression of the key transcription factor T-bet m RNA, and can delay the drift from Thl cells to Th2 cells of HIV infected people in some extent.2 Yiaikang capsule has the trend of improving the relative content of th22 cells and the relative expression of key transcription factor Aryl Hydrocarbon Receptors(Ah R) in the peripheral blood. This is probably the mechanism of Yiaikang capsule exerts effect on HIV infected people.
