Analysis The Core Prescriptions And Herbs Of Ulcerative Colitis By Data Mining And Study On The Influence Of Sophora, Rubia On Cellular Inflammatory Cytokines

Objective:To deal with information processing about Ulcerative colitis(UC) outpatient cases by using data mining methods, mining the UC’s core prescriptions drugs and compatibility rules could provide important data and information guide for the Chinese medicine treatment of this disease. To observe intestinal mucosa pathological changes of rats with ulcerative colitis by feeding Sophora and Rubia and determine the cell inflammatory factors on the experimental study, exploring possible mechanism by treating UC in these two traditional Chinese medicines.Methods:Clinical Study:Ulcerative colitis disease specialist outpatient data was collected to treat in structure. Importing the general condition, symptoms, tongue and pulse, syndrome, prescription, herbs and other data information into PL/SQL Developer. SPSS software was used to data in frequency statistics. Used Liquorice system to build complex networks to find core prescriptions and compatibility in the treatment of UC. Experimental Study:SD rats were randomly divided into normal group, model group, Sophora group, Rubia group and SASP group. Used TNBS/ethanol to induce the formation of UC. Rats were treated with Sophora, Rubia and SASP in 10 days. Collected serum and colonic mucosa samples of rats to observe pathological changes of colonic mucosa under light microscope. Then determinated the serum IL-6, IL-10, TNF-α by ELISA.Results:Clinical Study:Overall core prescriptions for the treatment of UC are Shaoyao Tang, Gegen Qin Lian Tang, Xianglian Wan, Baitouweng Tang. Large intestine damp-heat syndrome core prescriptions are Shaoyao Tang, Gegen Qin Lian Tang, Baitouweng Tang, Xianglian Wan and Huaihua San. Dame abundance due to spleen asthenia syndrome core prescriptions was Shen Ling Baizhu San, Sijunzi Tang. Liver-stagnation and spleen-deficiency syndrome core prescri-ptions was Tongxie Yaofang, Chaihu Shugan San. Yang deficiency of spleen and kidney syndrome core prescriptions are Lizhong Tang, Sishen Wan, Zhenren Yangzang Tang. Intertwist of cold with heat syndrome core prescription is Wumei Wan. Deficiency of yin and blood syndrome core prescription is Zhuche Wan. Core oral medicine are atractylodes, white peony root, coptis chinensis, radix auckladiae, angelica sinensis, tangerine peel, glycyrrhiza, garden buenet, agrimony, coix, scutellaria, chinese yam, poria cocos, medicated lesven, lithospermum, et al. There are 29 core compatibility like coptis chinensis-radix auckladiae, coptis chinensis-white peony root, coptis chinensis-garden buenet, white peony root-radix auckladiae, white peony root-garden buenet. Core medicine are garden buenet, salep, agrimony, lithospermum, phellodendron, panax notoginseng, glycyrrhiza, sophora, patrinia, glassleaved sweetflag rhizome, rubia, coix, sophora flower, ampelopsis japonica, scutellaria, white peony root, et al. There are 21 enema core compatibility like phellodendron-salep, phellodendron-garden buenet, salep-garden buenet, glassleaved sweetflag rhizome-garden buenet, phellodendron-glassleaved sweetflag rhizome. Experimental study:Sophora group and Rubia group rat colon gross morphological level and pathological scores were significantly improvement. Compared with the normal group, model group IL-6 and TNF-α were higher otherwise IL-10 was lower. Compared with model group, Sophora group, Rubia group IL-6, TNF-α levels were significantly lower (P<0.01). Sophora group IL-10 was significantly higher than the model group (P<0.01).Conclusion:This study figured out core prescriptions and compatibility by DM. Oral medicine often used to qing enteric and dissipate dampness, harmonize the qi and blood, strengthen spleen. Enema medication often used to dissipate heat and dry the damp, activate blood and absorb clots, Sore myogenic.All of these results fully reflected the Chinese understanding of UC, reveal the treatment path and medication characteristics. From an objective point of view it reflected the general rules and characteristics of prescription and medcine selection in the traditional Chinese medicine treatment of UC, which conformed to the previous knowledge and clinical practice understanding system. Data mining had application value by better feasibility study of TCM compound compatibility. Experimental study showed that Sophora, Rubia could repaire the mucosa of UC rats, so as to achieve the purpose of the treatment of ulcerative colitis by the possible mechanisms to reduces IL-6, TNF-α levels and increased level of IL-10.
