Objective:to research the change of CD8+CD28-Regulatory T cell expression in the patients of Primary Hepatocellular Carcinoma,and to explore the role which CD8+CD28- Regulatory T cell play in immune response.Method: Firstly,to test and count the CD8+CD28- Regulatory T cell in the peripheral blood of 60 cases of PHC and 50 cases healthy controls by flow cytometry technology,and then To test the CD8+CD28- Regulatory T cell in the Carcinoma tissue and Para- Carcinoma tissue of 46 surgical specimens by flow cytometry technology, lastly to test the CD8+CD28- Regulatory T cell in the Carcinoma tissue and Para- Carcinoma tissue of 30 paraffin specimens by immunohistochemical method.Result:By the contrast between the PHC cases and healthy controls,we found that CD8+CD28- Regulatory T cell was obviously increasing in PHC cases(21.02±3.14)% than healthy controls(15.12±3.16)%(P<0.05);Among the PHC cases,according to the size of the carcinoma,CD8+CD28- Regulatory T cell was obviously increasing in big size(21.83±2.71)% and huge size(23.61±3.83)% PHC cases than micro size(19.03±3.74)% and small size(18.79±2.28)% PHC cases,difference between big size and huge size was not obvious, difference between small size and micro size was not obvious,too. In the surgical specimens, the level of CD8+CD28- Regulatory T cell expression in the Carcinoma tissue(19.34±3.12)% was significantly higher than in the Para- Carcinoma tissue(13.59±2.47)%;In the paraffin specimens, the level of CD8+CD28- Regulatory T cell expression in the Para-Carcinoma tissue(9.47±3.04)% was significantly lower than in the Carcinoma tissue(13.58±2.46)%。Conclusion:Expression of CD8+CD28- Regulatory T cell was much more activated in the microenvironment of PHC cases,and maybe it plays an important role in immune response to PHC.
