[Objiective] To investigate the protective effects of deep hypothermic at subcellular level by research rat hippocampus mitochondrial permeability transition pore opening and closing at whole body Deep hypothermic circulatory arrest.[Methods] Thirty-three matured Sprague-Dawley(SD) rats, Three of them were used as blood-supply animal and the other thirty rats were divided randomly into three groups:deep hypothermic circulatory arrest(DHCA)group, normal temperature circulatory arrest(NTCA) group and normal rats control(NC) group. The closed-chest cardiopulmonary bypass(CPB) animal model was established in SD rats by means of external cannulation.In DHCA group circulatory arrest were performed in the temperature of deep hypothermia(< 27 ℃), for 10 minutes; In NTCA group circulatory arrest were performed in normal temperature (36.8℃~37.0℃), for 10 minutes; In NC group without circulatory arrest and deep hypothermia, The cardiopulmonary bypass time was 20 minutes. Rats’ hippocampus were removed from its brain and divided into two parts at the end of each courses. one part of the hippocampus was used to made frozen section and the other one was used to extract hippocampus mitochondria.use the way of immunofluorescence and western blot to measure mitochondrial permeability transition pore opening and closing status. Data were analyzed by software SPSS.[Results] There were more mitochondrial permeability transition pore open in NTCA group than in NC group (P<0.05).compared with DHCA group NTCA group’ s mitochondrial permeability transition pore opened more (P<0.05). DHCA group’s mitochondrial permeability transition pore opened more than NC group’s (P<0.05).[Conclusion] Ischemia hypoxia can lead to mitochondrial permeability transition pore opening; Deep hypothermia can inhibit mitochondrial permeability transition pore opening; Deep hypothermia by inhibiting the mitochondrial permeability transition pore opening to maintain the structure and function of mitochondria complete, ensure the normal metabolism of cells activity which have the effect of Cerebral protection. |
