[OBJECTIVE] To investigate the distribution pattern and efficacy of umbilical cord mesenchymal stem cells transplantation on MRL/lpr mice. Whether Huc-MSC can reduce the immune response, improve immune damage, discuss the influences of MSC on lupus nephritis.[METHODS] Twenty four 22-weeks-old MRL/lpr female mice were divided into 2 groups which are Poly-L-lysine hydrobromide-superpararnagnetic iron oxide-HUC-MSCs (PLL-SPIO-MSC) group (PG) and negative control group (CG). 1×106 PLL-SPIO-MSC were injected into per mouse with the PG. The control group was treated with 0.5ml normal saline as controls. After transplanting the HUC-MSCs into mice, we scaned both groups kidneys with MRI to observe the distribution pattern of HUC-MSCs. Testing the quantity of SRY gene in blood and tissue of mice by PCR. To observe the change of renal pathology, the weight, level of proteinuria among the different time of mice (before magnetic marker stem cells transplantation, instantly transplant, and 1day,3 days,5 days,8 days after transplantation). The levels of ANA, ds-DNA, Clq tested by enzyme linked immunosorbent assay (ELISA). The levels of IL-4, IL-6, IL-10, IL-17 and TNF-γ tested by the flow cytometry, and the results were recorded and analyzed.[RESRULTS] 1. There were no significant image change on MRI scan kidneys areas in both groups on each time point (before magnetic marker stem cells transplantation, instantly transplant,1 day,3 days,5 days and 8 days).2. No SRY gene was detected of PG mice on 3 days,5 days and 8 days after transplantation.3. After three weeks later transplantation, the weight of PG mice was significant higher than that of CG mice (P<0.05). The level of proteinuria in PC (0.39±0.13mg) was lower than that in CG (0.61±0.13mg) (P<0.05). PG’s kidney pathological changes slight than those in CG, immunofluorescence of C3 and IgA were not seen.4. The titer of ANA and ds-DNA in PG were significantly lower than the titer in CG (P<0.05). The concentration of Clq in PG (411±207mg/ml) was lower than that in CG (485±209mg/ml)(P>0.05).5. The concentration of IFN-gama in PG was higher than the concentration in CG (P<0.05). The levels of IL-17 and IL-10 in PG were lower than the concentration in CG (P<0.05).[CONCLUSIONS] The HUC-MSCs has some positive effect on MRL/lpr mice and it maybe inhibit immune system by reducing the levels of inflammation factors. |
