In this paper we review detaily the study on the chemical constituents and pharmacological activities of iridoid of rubiaceae plant for nearly a decade. On the basis of study of the extract of Saprosma merrillii Lo. has significant tumor inhibition of proliferation activity, we further study on its chemical composition. The air dried Saprosma merrillii Lo. are crushed, and then extracted by 75% ethanol (in volume fraction) at room temperature 3 times, merge the filtrate, recycled solvent, total ethanol extract. The extract was mixed with water, extracted by petroleum ether, and ethyl acetate, respectively. By petroleum ether, ethyl acetate and water. The three fractions were respectively subjected to normal and reversed-phase silica gel, recrystallization repeatedly and column chromatography over macroporous resin to obtain pure compounds. The chemical structures were established on the basis of extensive spectroscopic data analysis and by the comparison with spectroscopic data reported in the literature. Bioassay-guided isolation studies on the stem extract of Saprosma merrillii Lo. led to the isolation of thirty-five compounds. Compounds 1-3 were new triterpenoid. Compounds 1-3,7-19,21-22,24-35 were firstly isolated from this plant, except for compounds 4-5,20, 23 were firstly isolated from this genus.EtOAc soluable fraction:24 compounds were identified:(3R,6S,7R,8R,10R,19R,20R)-3a, 6A7A29-tetrahydroxylupan-28-oic acid (1), (3R,6S,8R,10R,19R,20R)-3a,6β,29-trihydroxy lupan-28-oic acid (2), (3R,8R,10R,19R,20R)-3a,29-dihydroxy-6-oxo-lupan-28-oic acid (3), lupeol (4), betulinic acid (5), ursolic acid (6), damnacanthol (7),3-hydroxy-l-methoxy-2-carbomethoxyanthraquinone (8),6-hydroxy-l-methoxy-2-methylanthraquinone (9), 2-hydroxy-3-methylanthraquinone (10),3-hydroxy-1,2-dimethoxyanthraquinone (11), 2-hydroxy-l-methoxyanthraquinone (12),1,3-dihydroxy-2-methylanthraquinone (13), 1,6-dihydroxy-2-methylanthraquinone (14),1,3,6-trihydroxy-2-methylanthraquinone (15), 3-hydroxy-l-methoxy-2-methyl etheranthraquinone (16), genistein (17), Daidzein (18), 5,7,2’-trihydroxy-4-methoxy-isoflavones (19),4,5,7-trihydroxyisoflavones (20), eupatolide (21), scopoletin (22), trans-Cinnamic acid (23), vanillic acid (24); Water soluable fraction: eleven compounds were identified:asperuloside (25), scandoside (26), 6"-O-trans-caffeoylgenipin gentiobioside (27), paederosidic acid (28), paederosidic acid methyl ester (29), paederosidic (30), mudanoside (31), clemochinenoside (32), clemahexapetoside A (33),3-O-β-primeveroside (34), kaempferol-3-O-β-D-glucopyranoside (35).The new compound 3 had the moderate activity against A549 cell with IC50 values of 12.61 μg/mL. The tests showed that compound 2,12 exhibited activity against E. coli with MIC values of 1.25 μg/taL. In addition, we also established the method of content determination of S. merrillii Lo stems of 28.
