| Objective:Primary liver cancer is a globle health threat as HCC (hepatocellular carcinoma,HCC) accounts for more than 90% in liver cancer. Radical operation,vascular intervention and liver transplantation are main therapy approach while their curative effect are not satisfying. Early studies had demonstrated that detection of tumor biomarker can predict the prognosis and optimize therapy. Mutant type of p53 and β2 receptor are related with survival and metastasis respectively. This study analyzed the expression of mutant type of p53 and β2 receptor,their correlation, and their relation with survival and metastasis tendency of HCC patients, therefore provide experimental basis for predicting the prognosis of liver cancer.Methods:The subject investigated were the clinical data and paraffin blocks of surgical resection specimens of 124 patients who were collected from January 2003 to December 2013 in the First Affiliated hospital of Dalian Medical University. Their histopathological type is hepatocellular carcinoma. The survival time of patients entering the research were inquired by telephone follow-up, out-patient review, Center for Disease Control and hospital medical record room. Using immunohistochemical Max Vision method to detect the expression of mutant type of p53 and β2 receptor, The numerical data is analyzed by Chi-square test, Spearman test analyze the correlation, Kaplan-Meier method was used in evaluating survival rate, Binary logistic was used to analyze the relation with survival and metastasis tendency.Resultsl.The expression of mutant type of p53 and 02 receptor in HCCThe number of positive expression of mutant type of p53 was 52 while negative expression was 72.The positive rate was 41.93%. The cases of positive expression of β2 receptor were 77 while negative expression was 47.The positive rate was 62.09%.2.The relation of clinical characteristics with mutant type of p53 and β2 receptorThere was no prominent relationship between mutant type of p53 expression and patients’ ages, genders, hepatitis infection history, AFP, tumor sizes, liver cirrhosis, (P>0.05).While there was correlation between mutant type of p53 and metastasis tendency(P<0.05). Ages, genders, hepatitis infection history, tumor sizes, liver cirrhosis showed negative association while AFP and metastasis tendency showed positive association with β2 receptor(P<0.05).3. The correlation of mutant type of p53 and β2 receptor, and the correlation’s impact on survivalAmong the 124 cases of HCC, the cases of simultaneous positive and negative expression of mutant type of p53 and β2 receptor were 35(28.22%) and 30(24.20%) respectively. The cases of positive expression of mutant type of p53 while negative expression of β2 receptor were 17(13.71%); the positive expression of β2 receptor while negative expression of mutant type of p53 were 42(33.87%). The Spearman test showed no obvious correlation between mutant type of p53 and β2 receptor(P=0.309).The survival analysis of these groups showed that simultaneous positive group was apparently lower than the simultaneous negative group. At the meantime, the survival of one positive group was median in simultaneous negative and positive group.4. The relation of simultaneous positive expression with survival and metastasis tendencyThe positive expression simultaneously of mutant type of p53 and β2 receptor has significant relation with metastasis tendency. The metastatic risk of positive expression simultaneously is 3 times,2.75 times,1.81 times than simultaneous negative group〠mutant type of p53 positive group and β2 receptor positive group respectively.ConclusionAmong patients with hepatocellular carcinoma after radical liver resection, the ones with simultaneous positive expression of mutant type of p53 and β2 receptor have shorter survival time than those with single positive expression respectively. The ones with simultaneous positive expression of mutant type of p53 and β2 receptor have higher metastatic risk than those with single positive expression. |
