| Objective:The purpose of this study is to investigate the expression of Histone methylase SET8in renal cell carcinoma,reveal the Relationship with the Wnt signaling pathway,and to explore its mechanism of action in renal cell carcinoma and its clinical significanceMethod:We randomly selected20patients from department of Pathology of Nanjing General Hospital of Nanjing Military Command between2010and2012who had accepted radical nephrectomy to analyze retrospectively.The pathological specimens are pathological diagnosis of renal cell carcinoma.All the specimens sliced, HE staining and then being confirm the diagnosis with light microscopy, there are18cases of renal cell carcinoma, including two cases with metastasis, two cases of collecting duct carcinoma with metastasis. We collected20Corresponding normal renal tissues about3cm away from the tumor as experimental control. All the patients were not receiving chemotherapy, radiation or other anti-tumor treatment before. According to the Preoperative examination, pathological diagnosis, re-pathological staging base on the2010AJCC staging system (7th ed.), re-pathological classification according to the2004WHO classification of renal cell carcinoma, tumor cell classification base on three classification of tumor cell grade. All the specimens borrowed department of Pathology of Nanjing General Hospital of Nanjing Military Command, Re-slicing the corresponding specimens and diagnosed as RCC or normal kidney tissue by HE staining. All the renal cell carcinoma and the corresponding normal kidney tissue were investigated by Immunohistochemical EliVision two-step staining with SET8and β-catenin. The stained slices were observed by200magnification of10mesh microscope.We used SPSS19.0software to statistical analysis of the experimental results, analyse the differences in expression levels of SET8and β-catenin. Then we analysis the relationship between the expression levels of SET8,β-catenin and the patient clinical information, pathologic stage, and tumor grading, and discuss the correlation of SET8and β-catenin protein expression in RCC.Results:Immunohistochemical staining of20cases of RCC specimens,SET8was positively stained in15cases,According to the WHO histological subtype classification:CCRCC13cases, CDC2cases;According to the TNM stage:Stage â… ,there are8cases; Stage â…¡,there are2cases;StageIII,there are2cases;Stageâ…¢,there are8cases.According to the tumor cell grade:G1, there are8cases; G2, there are4cases;G3, there are8cases.Statistical analysis, The Positive expression and staining intensity of SET8is associated with the Pathological TNM stage and the tumor cell grade of RCC(P<0.05). It has nothing to do with the clinical data (P>0.05).Immunohistochemical staining of20cases of RCC specimens,β-catenin was aberrant expression in14cases.According to the WHO histological subtype classification:CCRCC, there are12cases;CDC,there are2cases.According to the TNM stage:Stage â… , there are7cases; Stage â…¡, there are1cases;Stageâ…¢, there are2cases;Stageâ…£, there are4cases.According to the tumor cell grade:G1, there are8cases; G2, there are3cases;G3, there are7cases.The aberrant expression and staining intensity of β-catenin is associated with the Pathological TNM stage and tumor cell grade(P<0.05). It has nothing to do with the clinical data (P>0.05).Through statistical analysis we found, in renal cell carcinoma, SET8’s positive expression is related to the expression of β-catenin protein, and there is positive correlation between The positive expression of SET8and the abnormal expression of β-catenin protein(r=0.473, P<0.001).Conclusion:1. Histone methylase SET8mainly express in the the cytoplasm of kidney tissue and RCC tissue, partially in the cell membrane and nucleus.β-catenin protein mainly express at the cell membrane of renal tubular epithelial cells.2. The expression of SET8in RCC is closely related to the TNM stage of RCC and the tumor cell grade of RCC.3. In RCC, SET8has a high percentage of positive expression, β-catenin appears membrane negative expression or ectopic expression.4. In RCC, the positive expression of SET8is correlated with the abnormal expression of β-catenin. |
