| Objective:we evaluate the change of estrogen effect correlators before and after the treatment, and study the distinction between the different groups and between ectopia and positional endometrium, in order to obtain the possible target point of the therapy of expelling dampness and dissipating blood stasis in the treatment of adenomysis.Methods:we firstly established animal model of adenomysis by the method of transplating the pituitary gland of male mice into the uterus of the female mice, then we devided the adenomysis mice into several groups:high dose of Neiyikangfupian(NYKFP) group, medium dose of NYKFP group, low dose of NYKFP group; Danefukangjiangao(DEFKJG) group; Gestrinone group;3months group and6months group. We also had sham-operation group and blank group. Feeding by physiological saline for3months, we firstly executed the sham-operation group, blank group and3months group during the oestrus or anoestrus. While other groups healed by different medicine of group names, and6months group still fed by physiological saline for another3months, then we executed all groups during the oestrus or anoestrus. We observed and analysed the endometrum glands and stroma invasion degree of muscular layer by pathological section and HE stain, and we surveyed the change of P450arom〠COX2ã€PGE2by the technique of immunohistochemisty.Results:1.We observe Ectopic tissues on muscular infiltration and evaluate the degree of adenomyosis severity, normal group and sham-operation group is0, the oestrus of3months model group is2.67, while anoestrus is2.17, the oestrus of6months model group is3.50, while anoestrus is2.83, the degree indicates that3months group endometrial stromal cells and glands infiltrate muscles layer,6months group endometrial stromal cells and glands infiltrate inside and outside the muscle layer, which shows that both groups molding success, and disease severity increases with the course of the disease, operation process of building had no effect. 2.P450COX-2PGE2:in the uterus endomembrane, the the value of p450gradually increase after the construction of mice model of AM,6months group is higher than3months group,6months group and3months group are notably higher than normal group and sham-operation group, the compare of normal group and sham-operation group has no statistic signification. Compared with6months group, the indicates are all regulated down after treatment, NYKFP high dose group has notably statistic significance, DEFKJG group and gestrinone group compare with6months group has statistic significance. The compared between groups has statistic significance.Conclusions:1. We observe Ectopic tissues on muscular infiltration and evaluate the degree score of adenomyosis severity, the degree shows that both groups molding success, and disease severity increases with the course of the disease, operation process of building had no effect. The therapy of expelling dampness and dissipating blood stasis can regulate down the correlation factors of estrogen effect expression, interdict positive feedbackloop, restrain estrogen production to treat AM. So we can say P450ã€COX-2〠PGE2are the therapeutic target spots.3. NYKFP high dose group has a signification influence to the correlation factors of estrogen effect P450, cox-2, PGE2, which is superior to the other each treatment group, traditional Chinese medicine, not only effectively improve the abnormal effect of estrogen but also reduce the side effects of western medicine and reduce the cost, so it can bring higher cost performance to patients, it is worth in-depth study of traditional Chinese medicine compound.4. The correlation factors of estrogen effect expression is changed by estrous cycle, and the change has statistical significance. |
