| Objective:We aim to investigate miR-101fucnctions on U87, U251glioma cell lines,and to further study affect of miR-101on the the glioma cell lines mechanism theoretical and lay the foundation.Methods:U87, U251glioma cell lines were transfected by synthetic miR-101, and after transfection these cells were observed at different time points to observation the changes of cell proliferation,cycle,apoptosis and invasion, and also carried on statistics analysis. U87and U251cells were cultured by10%newborn calf serum, cells in logarithmic growth phase were divided into blank control group, negative control group, positive intervention group;MiR-101was Chemically synthesized, and U87and U251cells were transfected by transient transfection; The changes of cell proliferation at different time points to detection was evaluated by MTT assay;The changes of the cell cycle and apoptosis that at different time points were detected by flow cytometry;Also,transwell experiment and cell scratch test were use to detect malignant phenotype changes and analyze miR-101impact on glioma cell line function. All date are expressed as mean±SEM.(x±s).Statistical analysis was done by using SPSS13.0, by α=0.05for the inspection standard,to compare each group by ANOV.With P<0.05for significant differences.Results:MiR-101Overexpressed in U87and U251cells could reduce migration,inhibited at G0/G1phase cell growth,induce apoptosis significantly increased, suppress invasion.Statistical analysis showed that there was no statistical difference between the inhibitory effect of miR-101on U87of glioma cells and miR-101on U251of glioma cells.Conclusions:MiR-101is a tumor suppressor gene in glioma, miR-101overexpressied in U87cells and U251cells could inhibited the function of these glioma cells; MiR-101overexpressied in U87cells and U251cells could inhibition proliferation of glioma cell,inhibited at G0/G1phase cell growth,induce apoptosis significantly increased, and suppress invasion and reduce migration ability. |
