| Objectives:To explore the relationship between the effect of sinomenine on AIA pattern recognition receptors TLR8and RA treatment, and provide laboratory evidence for the clinical application of sinomenine in treatment of rheumatoidarthritis.Methods:SD rat paw of rats injected subcutaneously complete Freund’s adjuvant to establish palm AIA model. Sinomenine randomly divided into high, midium and low treatment group, positive control group and blank control group, model control group. Different methods of intervention in each group, then at Od,14d,21d,28d,35d,42day measurement of body weight, ankle circumference, foot pad thickness. Rats were sacrificed on day42inflammation-related indicators RF, CRP, IL-12, IFN-y, IL-10was prepared serum determination. Immunohistochemistry was used to detect peripheral immune organs spleen TLR8expression rate, data analysis and processing applications SPSS18.0.Results:(1) Sinomenine can effectively inhibit the body weight growth slowed in rheumatoid arthritis rats.(2) The Sinomenine can effectively reduce ankle swelling in rats.(3) Sinomenine can inhibit inflammation-related targets in rat serum RF, expression of CRP, IL-12, IFN-y, and promote the expression of IL-10.(4) Sinomenine spleen tissue can inhibit the expression of TLR8of pattern recognition receptors.Conclusions:Sinomenine by inhibiting expression of spleen tissue pattern recognit ion receptor TLR8,the TLR8signal channel conduction is suppressed, blockingthe activation of DC cells, so that the proinflammatory cytokines IL-12, IFN-y, cytokin e secretion and reduced the production, increase the anti-inflammatory cytokine I L-10cytokine production and secretion, regulate Thl/Th2balance, the inflammato ry reaction and bone destruction is suppressed and delay, so as to achieve the eff ect of anti rheumatoid arthritis. |
